Ümit ERDEMLİ Bu çalışma, Karakaya (Malatya) Baraj Gölünde bulunan ve ekonomik öneme sahip balıklar üzerinde, aylık periyotlarda eşey, yaş, boy ve ağırlık ilişkisine bağlı olarak kan hücrelerinin ortalama değerlerinin belirlenmesi amacıyla hematolojik yönden yapılmıştır. Aralık 1998 ile Aralık 1999 tarihleri arasında aylık periyotlar halinde sürmüş olan bu çalışma da baraj gölü üzerinde belirlenmiş (Morhamam, Hasırcılar, Boran ve Kömürhan Köprüsü Mevkii) dört istasyondan çeşitli ebatlarda fanyalı ve fanyasız ağlarla balıklar yakalanmıştır. Balıklar, önce oluşabilecek stresten uzaklaştırabilmek için içerisinde aynı göl suyu bulunan 40-50lt'lik plastik bidonlara alındı. Sonra kan almada"Canlı iken Kuyruk Kesme" yöntemi kullanılarak, kanın pıhtılaşmaması için direkt olarak EDTA'lı cam tüplere alındı. Balığın vücut sıcaklığı ile suyun sıcaklığı eşit düşünülerek, dış ortamla sıcaklık farkı nedeniyle alınan kan örneklerinin hemolize olmaması için, su sıcaklığındaki özel seyyar kaplara alındı ve çalışma süresince muhafaza edildi. Bu arada, ortamın su kalite kriterlerini tespit etmek için, 1.5-21t kadar su örneği alındı. Ayrıca, balığın taksonomik ve biyolojik (Yaş, Boy ve Ağırlık) özellikleri kaydedilerek, karınları açıldı, cinsiyet durumu ile parazitli olup oj, morfolojik olarak gözlendi. Ancak, parazitli örnekler hematolojik incelemeye alınn#ac$fc fi î|iV tM v- - ->~- -.V..5ı. -5 Muayenede lmm kanda; eritrosit ve trombosit sayısı, hemoglobin miktarı, hematokrit değer, eritrosit indeksleri (MCV, MCH ve MCHC) ve total lökosit sayısı belirlenerek, lökosit tiplerinin (Granüler ve Agranüler Lökositler) yüzdedeki oranlarını belirlemek amacıyla kan frotileri de hazırlandı. Ayrıca, eritrosit hücrelerinin yıllık periyottaki çap büyüklükleri cinsiyet farkına dayalı olarak belirlendi. Hematolojik analizlerden elde edilen bulgular ortalama ve standart hataları ile birlikte yaş-boy-ağırlık ve eşey-aylar şeklinde yıllık dağılımları tablolar halinde verildi. Tablolardaki veriler aynı şekilde grafiksel olarak da değerlendirildi. Hematolojik incelemede, erkek bireylerin genellikle dişilerden daha yüksek kan değerlerine sahip oldukları belirlenerek, bu parametrelerden özellikle, eritrosit sayısı, hemoglobin miktarı ve hematokrit değerler arasında yüksek bir korelasyon olduğu görüldü. Bulgularda, eritrosit sayısı ile su sıcaklığı arasında doğrusal bir ilişki görülürken, suyun oksijen konsantrasyonu (BOD) ile ters bir ilişkide olduğu saptandı. Balık türüne bağlı olarak üreme zamanlarına denk gelen dönemlerde özellikle nötrofil ve hematokrit değerlerde artış gözlendi. Ayrıca beslenme, çevrenin fiziksel ve kimyasal özelliklerine bağlı olarak hematolojik parametrelerde dalgalanmalar görülmesine rağmen, yıllık periyotta, kan hücre tiplerinin ilkbahar ve yaz dönemlerinde arttığı, sonbahar ve kış dönemlerinde ise azaldığı tespit edildi. Çalışma sonunda, yaş-boy-ağırlık ilişkisine bağlı olarak yapılan istatistiksel analizlerde, hematolojik parametrelerden; nötrofil, lenfosit ve monosit yüzde oranlarının yaş bağımsız değişkenlerinden anlamlı düzeyde etkilendiği görüldü. Bununla birlikte, eritrosit, hemoglobin, hematokrit ve bunlara bağlı olarak ta eritrosit indekslerinin hem yaş ve hem de ağırlık bağımsız değişkenlerinden anlamlı düzeyde etkilendiği belirlendi. Ümit ERDEMLİ This study was carried out to determine the average values of blood cells according to sex, age, length and weight a haematological point of view on some fishes, which live at Karakaya Dam Lake and have economic importance. The study continued between December 1 998-December 1999 and samples of fish were collected both with net and without net on monthly periods from previously determined four different stations (Morhamam, Hasırcılar, Boran ve Kömürhan Köprüsü Mevkii). Firstly, samples were taken into plastic buckets having 40-50 It lake water in order to prevent the samples from a possible stress. Blood samples were taken with the method "cutting the tail when the fish is alive" and they were directly put into glass tubes with EDTA in order to prevent the bloodletting. The tubes were placed into specially designed movable plates filled with the lake water in order to prevent the blood samples becoming hemolised and the tubes were kept there during the process. Mear amount of l,5-21t water sample was taken to control the water quality of the e#y#ont *SS5g5=@gj$ IV Besides the taksonomik and biological characteristic of the samples were recorded, their stomachs were opened and they went through a morphological analysis to find out if they have parasites. The samples with parasites were not included into the hematologic examination. During the examination the number of erytrocyte and trombocyte, the amount of hemoglobin, the value of hematocrite, erytrocyte index (MCV, MCH and MCHC), and the number of total leucocyte were determined together with the percentages of leucocyte types (Granular and Agranular leucocytes). Furthermore, the size of eryrocyte cells was determined on yearly period based on the sex. The findings obtained the hematologic analyses were presented in yearly tables including averages and standard errors according to age, length, weight and sex on monthly basis. The data in the tables were also presented graphically. Haematologic analyses reveal that the male fish generally have higher blood values than the female ones, and there was a high correlation, especially, between the number of erytrocyte, the amount of hemoglobin and the values of hematokrite. The finds also show that although there was a linear reaction between the number of erytrocyt and the degree of water, there was an opposite relation in the oxygen concentration of water. The study shows that there was an increase in the values of notrophille and hematocrite, especially, in the periods of same time production in different types of fish. In addition it was observed that although there were undulations in haematologic parameters depending, on nourishment and the physical and chemical characteristic of the environment, the types blood cells increased during spring and summer periods and reduced an fall and winter. The results of the haematological analyses made depending an the age, length and weight reveal that the percentages of notrophille, lenfocyte and monocyte were affected by age, independent variable and by haematological parameters. Besides, erytrocyte, hemoglobine and eryrocyte indexes were also affected by age, weight and independent variables at a significant level.
