7. ÖZET Serbest oksijen radikalleri, iskemi/reperfuzyon sonrasındaki serebral hasarın patoj enezine önemli rol oynar. Bu çalışmada sıçan beyninde oluşturulan iskemi ve reperfuzyonun beyin dokusuna yaptığı hasar ve bu hasara antioksidan özellikli mukolitik bir ilaç olan erdosteinin etkilerini in vivo olarak incelemek amaçlanmıştır. 10 dakika karotid arterler bilateral oklüze edilerek iskemi ve 30 dakika resirkülasyon yapılarak reperruzyon gerçekleştirildi. Erkek Wistar Albino türü sıçanlar 4 grup olarak belirlendi. İlk grup: hiçbir ilaç verilmemiş olan kontrol grubu; 2. grup: 3 gün boyunca sadece oral erdostein tedavisi (10 mg kg"1 b.wt. day"1) uygulanan grup; 3. grup: bilateral karotid arter oklüzyonu yapılarak 10 dakika iskemi, ardından 30 dakika reperruzyon uygulanan grup; 4. grup: İ/R ile birlikte, bu işlem yapılmadan önce 3 gün boyunca erdostein uygulanan grup. Beyinde l/R grubunun MDA seviyesi, diğer gruplara göre daha yüksek bulundu. İ/R + erdostein grubunun glutatyon peroksidaz (GSH-Px) ve katalaz (CAT) aktivitesi, İ/R grubu ile karşılaştırıldığında arttığı gözlendi. Sonuçta, beyinde İ/R uygulanması, lipid peroksidasyonunun artışıyla sonuçlandı ve erdostein tedavisi, sıçanlarda özellikle GSH-Px ve CAT antioksidan enzimlerinin aktivitesini artırarak oksidatif hasan engellemede yardımcı olduğu değerlendirildi. 8. SUMMARY Oxygen-derived free radicals have been implicated in the pathogenesis of cerebral injury after ischaemia/reperfusion. The purpose of the present study was to investigate the effects of ischaemia and subsequent reperfusion on rat brain and to find out the effects of erdosteine, a mukolytic drug showing antioxidant properties, on this in vivo model of cerebral injury. Ischaemia was induced by bilateral occlusion of the carotid arteries for 10 min. and reperfusion was achieved by releasing the occlusion to restore the circulation for 30 min. Male Wistar Albino rats were divided into four groups. The first group (group 1) that underwent no medication was accepted as control group; the second group (group 2) was treated with oral erdostein alone (10 mg kg"1 b.wt. day"1 for 3 days); the third group (group 3) underwent 10 min of global ischaemia by bilateral carotid artery occlusion followed by 30 min of recirculation period; and in the last group (group 4) erdosteine was administered starting before I/R for 3 days. MDA level of I/R group was higher than those of the other groups in rat brain. The activities of glutathione peroxidase (GSH-Px) and catalase (CAT) were increased in I/R plus erdosteine group in comparison with I/R group. In conclusion, I/R administration resulted in increased lipid peroxidation in the brain and erdostein treatment helped to prevent oxidative injury by increasing antioxidant enzymes, especially GSH-Px and CAT in rats.
