ÖZET Doktora Tezi KRONİK ALKOL VE SİGARA KULLANIMINA MARUZ BIRAKILAN ERKEK RATLARIN TRAKEA, AORT, ÖZOFAGUS VE MİDE DOKULARININ HİSTOPATOLOJİK YÖNDEN ARAŞTIRILMASI Kenan ERDOĞAN İnönü Üniversitesi Fen Bilimleri Enstitüsü Biyoloji Anabilim Dalı 67+DC sayfa 2004 Danışman : Prof. Dr. Eşref YÜKSEL Bu tezde sigara ve alkol kullanımının; trakea,aort, özofagus ve mide dokularına etkilerinin incelenmesi amaçlanmıştır. Her birinde 7 adet erkek Wistar rat bulunan 4 grup (Kontrol, sigara, alkol ve sigara+alkol) oluşturulmuştur. Alkol grubundaki ratlar inek sütü, vitamin A (5000 İÜ) ve sükrozdan oluşan diyeti % 7,2'lik alkolle birlikte ağız yoluyla alırlarken, sigara ve sigara+alkol grubundaki ratlar bir plastik kafes (0,75x1x0,85 m, 0,64 m3) içerisine yerleştirilerek, günde 20 adet sigara dumanına maruz bırakıldılar. 6. ayın sonunda 4 gruptaki hayvanların dokuları alınarak fiksasyon, dehidrasyon ve parafinizasyon işlemleri sonucunda uygun kesit için bloklandı. Bloklardan 4 mikron kalınlığında kesitler alındıktan sonra deparafinizasyon ve Harris' hematoksilen&Eozin (HE), Periyodic Acid SchifF(PAS), Alcian Blue& Periyodic Acid Schiff (AB&PAS) ve Masson's trichrom boyası ile boyanarak ışık mikroskobu yardımıyla histopatolojik açıdan değerlendirildi. Mide dokusunda, bağ doku ve ephel dokuda yoğun infiltrasyon, metaplazi, submukoza tabakasında yağ doku birikimi gözlenirken, bu belirtiler sigara+alkol grubunda daha belirgindi. Özofagusta epitel dokuda incelme, lümene bakan kısımda keratin birikimi, lümene doğru uzanan girintilerin kaybolması gibi histopatolojik bulgular mevcuttu. Trakea da yapılan incelemeler sonucunda, submukoza ve mukozada infiltrasyon, mukozada yer yer doku kaybı ve sillerde düzleşme görüldü. Damar endotelini araştırmak için aorttan kesitler alındı. Elastik liflerde ve hücre sayısında azalma ile birlikte mukoza tabakasında bir incelme gözlendi. Ancak daha kesin sonuçların ortaya konabilmesi için elektron mikroskobu ve değişik boyama yöntemlerinin (immunohistokimyasal) kullanılması gerekmektedir. Sonuç olarak özofagus, trakea ve mide üzerinde gözlenen hipertrofi, metaplazi, infiltrasyon ve keratinizasyon sigara+alkol grubunda daha fazlaydı. SUMMARY Ph. D. Thesis A fflSTOPATOLOGICAL INVESTIGATION ON THE TISSUES OF TRACHEA, AORTA, ESOPHAGUS AND STOMACH OF MALE RATS TREATED WITH CHRONIC ALCOHOL AND CIGARETTE SMOKE Kenan ERDO?AN İnönü University Graduate School of Natural and Applied Sciences Department of Biology 67+LX pages 2004 Supervisor : ProfDr. Eşref YÜKSEL The aim of the study carried out in this thesis was to investigate the effect of chronic cigarette smoke and alcohol exposure on the trachea, aorta, esophagus and stomach. Animals, treated with these substances for a period of six months were divided into four experimental groups which were i) a control group ii) alcohol group iii) cigarette smoke group iv) smoke+alcohol group. All groups were consisted of 7 rats. Animals of alcohol group were orally fed on a diet containing cow milk, 5000 IU vitamine A and sucrose together with 7.2% alcohol. Cigarette smoke group on the other hand were kept in closed plastic cages (0,75x1x0,85m, which makes 0,64 m3) and exposed to cigarette smoke for the whole experimental period, (20 cigarette per day). At the end of the experimental period, the speciments in four group were sacrified and then their relevant tissues were blocked for sectioning. The blocked tissues sectioned at 4 u tikness and deparafinized and dyed with Harris' Hematoksilen&Eozin (HE), Periodic Acid Shiff (PAS), Alcian Blue& Periodic Acid Shiff (AB&PAS) and Masson's HI trichrome. The preparations were examined histopatologically with an aid of light- microscope. In the results, dense inflammation and metaplazi were observed in connective tissues and epitelium of stomach, together with accumulation of fatty tissue at submucosa layers. This was most apperant in cigarette-alcohol group. In the esophagus, a reduction in the epitelium keratinization at the inner side and disappearance of microvilly were observed. Examination of trachea showed that there were inflammation of mucosa and submucosa layers and also local tissue degeneration and flattening of cilia. Thinn sections taken from aorta showed that there were a reduction in the number of cells, and also a reduction in the elastic fibres under the endotehum. Mucosa layer was also thinner compared to the controls. However, we need more investigation using immunohistochemical methods and electron microscopy micrograps for more definite results. We concluded that cigarette+alcohol creates more inflammation, metaplazi and keratinization in trachea, esophagus and stomach. This shows that cigarette and alcohol may have sinergetic effect.
