Öz:Son yıllarda karsinogenez sürecinde oksidatif stresin önemli bir role sahip olduğuna dair bir hayli delil bulunmuştur. MCF-10A hücrelerinin (insan meme epitel hücreleri) bir tümör promotor olan 17-β-estradiol (E2) ve meme için karsinojen bir ajan olan 7,12-dimetilbenz(a)anthracene (DMBA)'e maruz bırakılması, reaktif oksijen türlerinin üretimi üzerinden hücrelerin neoplastik transformasyonunu etkileyebilir. Bu çalışmada MCF-10A hücrelerinde serbest radikal süpürücü enzim aktiviteleri üzerine E2 ve DMBA'nın etkisi araştırıldı. Bu amaçla hücre kültür ortamına 4, 8 ve 12 haftalık sürelerle 0 μM, 10 μM, 25 μM DMBA ve 0 M, 10-9 M ve 10-8 M E2 ilave edildi. Bu maruziyet periyodundan sonra hücreler triton X-100 ihtiva eden hipotonik solüsyonda parçalandı. Bu hücre ekstraktlarında süperoksit dismutaz (SOD), katalaz (CAT) ve glutatyon peroksidaz (GSH-Px) aktiviteleri belirlenerek birbiriyle karşılaştırıldı. 10 μM DMBA ve 10-9 M E2 ile 12 hafta süreyle muamele edilen hücrelerde GSH-Px aktivitesinde kontrol grubuna göre önemli bir artış tespit edildi (kontrol grubunu, DMBA grubu için %0.1 aseton ve E2 grubu için %0.01 etanol ile muamele edilmiş hücreler oluşturdu). SOD ve CAT aktiviteleri DMBA ve E2'nin konsantrasyonlarına ve maruziyet zamanına göre irregüler bir değişim gösterdi.
Abstract:There is a great evidence within the past few years that oxidative stress has an important role in the process of carcinogenesis. Exposure of MCF-10A cells (human breast epithelial cell line) to the mammary carcinogen 7,12-dimethylbenz (a)anthrecene (DMBA) and possible tumor promoter 17-β-estradiol (E2) may affect the neoplastic transformation of cells leading to production of reactive oxygen species (ROS). The effects of E2 and DMBA on free radical scavenging enzyme activities have been investigated in MCF-10A cell lines in this study. MCF-10 cells were treated with 0 μM, 10 μM, 25 μM DMBA, and 0 M, 10-9 M, 10-8 M E2 for 4, 8, and 12 weeks in cell culture media. After these exposure periods, cells were homogenized in hypotonic solution with Triton X-100. Superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT) activities were determined in these cell extracts and compared with each other. There is an increase in GSH-Px activity in cells treated with DMBA (10 μM) and E2 (10-9 M) for 12 weeks compared with control cells (treated with only acetone 0.1% and ethanol 0.01%, respectively, for 12 weeks). SOD and CAT activities were changed significantly in a irregular manner depending on concentrations of DMBA and E2, and exposure time.
