1 ÖZET. Çalışmamızın amacı; erkek D. melanogaster Oregon yabanıl tip ve D. melanogaster vestigial mutantının ömür uzunlukları farklılıklarını, serbest radikal temizleyicileri olan katalaz; glutatyon redüktaz aktiviteleri ; redükte ve okside glutatyon seviyelerindeki değişiklikleri yaşlanma süreci içerisinde incelemekti. Erkek Oregon D. melanogaster in ortalama ömürlü D. melanogaster vestigial mutantından daha uzundu. Oregon'da maksimum ortalama ömür 85 gün, vestigial 'de ise 56 gündü. Yaslanmaya bağlı olarak katalaz aktivitesindeki değişiklikler Oregon ve vestigial" de aynıydı ve her ikisinde de yaslanma süresince azalan bir eğim gösterdi. Oregon'da glutatyon redüktaz aktivitesi 10 - 40. günler arasında biraz arttı ve daha sonra hızla dUstll. Yaslanmaya bağlı olarak vestigial' de glutatyon redüktaz aktivitesi 40 güne kadar % 60'lık bir azalma ve ömrün son günlerinde belirgin artış ile kavisli bir eğim gözlendi. Erkek Oregon D. melanogaster 'de redükte glutatyon (GSH) ve okside glutatyon (GSSG) miktarı 40 gün boyunca biraz arttı, 40 günden sonra belirgin bir azalma gösterdi. Vestigial 'de GSH ve GSSG seviyeleri ilk 10 gün için değişmeden kaldı. 20 gün civarında belirgin bir azalma gösterdikten sonra arttı.
ABSTRACT. The objective of our study was to investigate the differences in the life span of male Oregon (w.t.) and vestigial Drosophila melanogaster, the possible role of free radical scavengers such as catalase, glutathione reductase and reduced - oxidized, glutathione levels in the aging process by studying the pattern of age - related changes. The life span of male Oregon D. melanogaster is longer than vestigial D. melanogaster. The maximum life span was 85 days in Oregon population and 56 days in vestigial population. '" Age - related changes of catalase activities were similar in male Oregon and vestigial and showed a dec reasing curve during the aging. Glutathione reductase activity of Oregon increased slightly between 10 and 40 days, decreased sharply thereafter. Age-related changes in glutathione reductase activity of vestigial followed a pot-shaped trend with 60 % decrease during the 40 days and followed by a sharp increase during the late part of life. The amounts of reduced glutathione (GSH) and oxidized glutathione (GSSG) increased slightly during 40 days and followed a sharp decline after 40th day in male Oregon D. melanogaster. In male vestigial D. melanogaster, the concentrations of GSH and GSSG remained quite stable for the first 10 days and followed by a sharp decline around 20th day and then increased thereafter.
