ÖZET: i Bu çalışmada, organofosforlu bir insektisit olan azinphosmethyl'in (Guthion) laboratuvar koşullarında, 28 hafta süre ile oral olarak 0.1 ppm ve 1 ppm' lik subletal dozlarda swiss albino fareler üzerindeki morfolojik, anatomik, histolojik ve biyokimyasal etkileri araştırılmıştır. Araştırma sonunda uygulama grubu farelerin aylık peryotlarla kaydedilen vücut ağırlıkları ve beslenme durumları kontrol grubu ile karşılaştırıldığında aralarındaki fark istatistiksel olarak önemli bulunmuştur. Anatomik incelemelerde, uygulama grubu farelerde iç organlar çevresinde yağlanmanın fazla, karaciğer ve böbreğin büyüklük ve renk açısından kontrol farelerden farklı olduğu gözlenmiştir. Histolojik incelemelerde, kullanılan pestisitin karaciğerde, böbrek ve beyinden daha fazla hasara yol açtığı tesbit edilmiştir. Uygulama grubu birinci ve ikinci nesil farelerin karaciğer dokularında, çift nukleuslu hepatositler, mitotik aktivitede artış, pleomorphism; Dokuda hepatosit zedelenmesine bağlı olarak fokal nekroz odaklan, kupffer hücrelerinin sayısında artış, sinozoidlerde genişleme ve konjesyonun meydana geldiği saptanmıştır. Biyokimyasal incelemelerde ise, Uygulama ve kontrol grubu farelerden gözden kan alma tekniği ile elde edilen plazma örneklerinde kolinesteraz (ChE) enzim aktivitesi spektrofotometrik olarak analiz edilmiştir. Plazma ChE aktivitesinin 1 ppm uygulama grubunda erkek farelerde %52.14ve dişilerde %4 1.29 oranında inhibe edildiği, 0.1 ppm uygulaması içinde bu oranlar sırası ile %31.0 ve %29.84 olduğu saptanmıştır. M Uygulama gruplarına ait dişi ve erkek hayvanlarda enzim aktiviteleri farkı istatistiksel olarak önemli bulunmuştur (P<0.001 ; P<0.02). ABSTRACT Lii In this study, morphological, anatomical, histological and biochemical effects of azinphosmethyl (an organophosphorus insecticide) at 0.1 ppm and 1 ppm sublethal dozages on swiss albino mice have been investigated for 28 weeks at laboratory counditions. In the results, the difference between mouthly recorded body weights and food consumption of mice under investigation and that of control group were found to be statistically important. In anatomical study, there were more fatty tissue around some internal organs such as liver, kidney in investigation groups than control group. These organs were also found to be different than in controls. in organ size and colour. It was found that the insecticide makes more damage in liver than brain and kidneys. There were not only double nucleated hepatocyt, increase in mitotic activity and pleomorphism in the first and second generations of investigation groups; and also focal necrosis sides caused by hepatocyt damage, an increase in the number of kupffer cells, enlargement of sinosoids and congestion were seen. In the biochemical analysis, cholinesterase (ChE) enzyme activities were measured spectrophotometrically from the plasm samples obtained from blood taken from the eyes.- Plasm ChE activity of 1 ppm group were inhibited at the rate of 52.14% and 41.29% in males and females respectively. The corresponding values in 0.1 ppm group were 31% and 29-28%. The difference in enzyme activity between males and females of investigation groups were found to be statistically significant.
