ERDEMOĞLU Bu çalışmada ilk olarak Türkiye'de tedavi amacıyla yaygın olarak kullanılan adaçayı, ısırgan otu, rezene, ıhlamur, papatya, senemaki ve kuşburnu örneklerinde ve bunların infüzyonlarındaki 14 adet makro ve mikro elementin (Ca, Mg, Mn, Fe, Cu, Zn, Al, Ba, Sr, Ni, Co, Cr, Cd, Pb) tayinleri yapıldı. Bu bitkilerin analizlerinden önce bitkilerin çözünürleştirme işlemleri mikrodalga ile parçalama ve yaş yakma teknikleri kullanılarak yapıldı. Bitkilerdeki ve infüzyonlarındaki Ca ve Mg derişimleri alevli atomik absorbsiyon spektroskopisi (FAAS) ile, Fe, Cu, Zn, Al, Ba, Mn, Sr, Ni, Co, Cr, Cd ve Pb içerikleri ise indüktif eşleşmiş plazma atomik emisyon spektroskopi (ICP-AES) teknikleri ile belirlendi. Doğruluk ve kesinliğin test edilmesi amacıyla GBW 07605 sertifikalı referans madde kullanıldı. Bu çalışmanın bir diğer konusu ise Türkiye'de alışılagelmiş biçimde tedavi amacıyla kullanılan bu bitkilerin infüzyonlarındaki gallik asit, epikateşin gallat, epigallokateşin, epigallokateşin gallat ve epikateşin polifenollerinin seviyelerini belirlemektir. Polifenollerin analizi için uygun bir örnek hazırlama basamağından (su içerisine ekstraksiyon) sonra her bir polifenolün eş zamanlı ayrılması, tanınması ve kantitasyonu için ekonomik bir hareketli faz ile basit, hızlı ve kesin olan gradient sistem ters faz-HPLC metodundan yararlanılmıştır. ERDEMOGLU In this study, fourteen macro and micro elements (Al, Ba, Ca, Cd, Co, Cr, Cu, Fe, Mg, Mn, Ni, Pb, Sr and Zn) were determined in the herbs that are widely consumed for medical purposes as tea in Turkey such as chamomile (Matricaria chammomile L.), fennel (Foeniculum vulgare), linden (Tilia vulgaris), nettle (Urtica dioical), rosehip (Fr.Rosa caninae), sage (Salvia officinalis) and senna (Cassia anqustifolia) and their infusions. Before the analysis of this herbs microwave digestion and wet digestion procedures were performed for dissolution of herbs. Ca and Mg concentrations in the herbs and infusions were determined by FAAS, Fe, Cu, Zn, Al, Ba, Mn, Sr, Ni, Co, Cr, Cd and Pb concentrations were determined by ICP-AES. The accuracy and precision were verified against a GBW 07605 Poplar leaves and Tea certified reference material. The other objective of this study is to establish the levels of polyphenols such as gallic acid, epicatechin gallate, epigallocatechin, epigallocatechin gallate and epicatechin in the infusions of this herbs habitually consumed for medical purposes in Turkey. After a suitable sample preparation procedure (water extraction) for polyphenols analysis, a simple, rapid, and precise gradient RP-HPLC method with an economical mobile phase was utilized for simultaneous separation, identification and quantification of individual polyphenols.
