Amaç: Çeşitli kanser hücrelerinde metilasyon duyarlı restriksiyon enzimlerinden ve RT-qPZR tekniğinden yararlanılarak protein katlanma, degredasyon ve otofaji yolaklarına ait bazı anahtar genlerin metilasyon profillerinin araştırılması ve hücreler arası epigenetik farklılıkların ortaya konması amaçlanmıştır. Materyal ve Metot: Hücreler kültür ortamına alıştırıldıktan sonra moleküler testler kullanılarak değerlendirilmiştir. Metilasyon spesifik restriksiyon enzimleri aracılığıyla (Acil-HpaII), DNA kesimi sağlanıp RT-PZR testleri ile metilasyon düzeyleri incelenmiştir. Bulgular: Otofaji yolağında rol alan genlerin (LRRK2, PRKN, GBA ve NBR1) promotor bölgelerinin metilasyon düzeyleri istatistiksel olarak karşılaştırıldı. GBA (P<0.05) geni hariç diğer genlerde anlamlı farklılık bulunamamıştır. GBA geninin promotor bölgesindeki metilasyon düzeyi, MCF 7 ve HEP-G2 hücrelerinde normal MCF 10A hücresine göre anlamlı düşük saptandı. Kanserli hücreler karşılaştırıldığında da anlamlı fark bulunmuştur. Endoplazmik Retikulum stres yolağında rol alan genlerin (MAP3K5, ATF6, XBP1, EIF2AK3 ve MAOA) promotor bölgelerinin metilasyon düzeyleri istatistiksel olarak karşılaştırıldı. Tüm genlerde HEP-G2 hücresi, normal MCF 10A hücresine göre istatistiksel olarak metilasyon düzeyinde azalma bulunurken; MCF 7 hücresinin normal MCF 10A hücresine göre sadece XBP1 ve EIF2AK3 genlerinde anlamlı artış bulunmuştur. Kanserli hücreler karşılaştırıldığında ise ATF6 ve XBP1 genlerinde anlamlı fark bulunmazken; MAP3K5, EIF2AK3 ve MAOA genlerinde anlamlı fark bulunmuştur. Sonuç: Otofaji yolağında rol alan genlerin metilasyon düzeyleri karşılaştırıldığında GBA geninde hem MCF 7 hem de MCF 10A hücrelerinde hipometilasyon saptanmıştır. Endoplazmik Retikulum yolağında ise tüm genlerin kanser hücrelerinde normal hücreye göre anlamlı şekilde azaldığı sadece MCF 7 hücrelerinde XBP1 ve EIF2AK3 genlerinde hipermetilasyon saptanmışır. Aim: It is aimed to investigate the methylation profiles of some key genes belonging to protein folding, degradation and autophagy pathways in various cancer cells and to reveal epigenetic differences between cells by using methylation sensitive restriction enzymes and RT-qPCR technique. Material and Method: After the cells were acclimated to the culture medium, they were evaluated using molecular tests. By means of methylation specific restriction enzymes (Acil-HpaII), DNA was cut and methylation levels were examined by RT-PCR tests. Results: The methylation levels of the promoter regions of genes (LRRK2, PRKN, GBA and NBR1) involved in the autophagy pathway were statistically compared. Except for the GBA gene (P<0.05), no significant difference was found in other genes. The methylation level in the promoter region of the GBA gene was found to be significantly lower in MCF 7 and HEP-G2 cells compared to normal MCF 10A cells. A significant difference was also found when cancer cells were compared. The methylation levels of the promoter regions of genes (MAP3K5, ATF6, XBP1, EIF2AK3 and MAOA) involved in the Endoplasmic Reticulum stress pathway were statistically compared. While there was a statistically decrease in methylation level in HEP-G2 cell compared to normal MCF 10A cell in all genes; Only XBP1 and EIF2AK3 genes were significantly increased in MCF 7 cell compared to normal MCF 10A cell. When cancer cells are compared, there was no significant difference in ATF6 and XBP1 genes; Significant differences were found in MAP3K5, EIF2AK3 and MAOA genes. Conclusion: When the methylation levels of the genes involved in the autophagy pathway were compared, hypomethylation was found in the GBA gene in both MCF 7 and MCF 10A cells. In the Endoplasmic Reticulum pathway, all genes were significantly decreased in cancer cells compared to normal cells, and hypermethylation was detected in XBP1 and EIF2AK3 genes only in MCF 7 cells.
