Öz:AMAÇ: Hidatik kistte canlılık tayini, in vitro ve in vivo çalışmaların yanı sıra ilaç denemelerine yönelik çalışmalarda da önem arz etmektedir. Fertil kistlerde protoskolekslerin canlılık oranlarını saptamak amacıyla neutral red, methylene blue, eosin, Papanicolao, Giemsa, Ziehl-Neelsen, toluidine blue ve trypan blue gibi çeşitli vital ve avital boyalar kullanılmaktadır. Yaygın olarak ise eozin tercih edilmektedir. Prosedürlerde farklı konsantrasyonlarda eozin kullanıldığı görülmektedir. Bu çalışma ile protoskoleks canlılığının araştırılmasında kullanılan eozinin ideal konsantrasyonunun belirlenmesi amaçlanmıştır. YÖNTEMLER: Protoskoleksler eozinin % 0.05, % 0.1, % 1, % 3 ve % 5‘lik konsantrasyonları ile muameleye tabi tutulmuştur. Eozin boyama yöntemi ile 500 protoskoleks canlılık yönünden 0., 5., 15., 30. ve 60. dakikalarda incelenmiştir. BULGULAR: Alev hücreleri motilitesi ve kontraktilitesinin değerlendirmesine dayalı protoskolekslerin canlılık testinde kullanılacak eozin için ideal konsantrasyonlar % 0.1 ve % 1 olarak saptanmıştır. % 1’den yüksek eozin konsantrasyonlarında; canlı protoskolekslerin koyu boyanan zemin üzerinde ayırt edilmesinin zor olduğu izlenmiştir. % 0.1’den düşük konsantrasyonlarda ise; alev hücresi motilitesi ve kontraktilite olmamasına karşın, ölü protoskolekslerin “canlı” olarak hatalı bildirilebileceği belirlenmiştir. SONUÇ: Eozin ucuz, toksisitesi az, kolay hazırlanabilir ve ulaşılabilir bir boya maddesidir. Bu özellikleri nedeniyle protoskolekslerin canlılık testlerinde tercih edilen eozinin, ideal boyanma kalitesi ve doğru canlılık kararına yardımcı olabilmesi için % 0.1 ve % 1’lik konsantrasyonların kullanılması önerilmektedir.
Abstract:OBJECTIVE: The viability assessment in hydatid cyst is important for both in vitro and in vivo studies, and also drug experiments. Several vital and avital stains as neutral red, methylene blue, eosin, Papanicolao, Giemsa, Ziehl-Neelsen, toluidine blue and trypan blue have been used to assess viability of protoscoleces in fertile cysts. Of them, eosin is widely preferred stain. Various concentrations of eosin have been used in the procedures. In this study, it was aimed to determine the ideal concentration of eosin for assessing protoscolex fertility. METHODS: Protoscoleces were treated with these eosin concentrations: 0.05 %, 0.1 %, 1 %, 3 %, and 5 %. A total of 500 protoscoleces were examined by eosin staining techniques with 0., 5., 15., 30. and 60.-minute intervals. RESULTS: Ideal concentrations of eosin which will be used for viability test to determine the motile flame cells and contractile protoscolex were found as 0.1 % and 1 %. In viability test of protoscolex, more than 1 % concentrations of eosin dying caused difficulty for the decision of viability because of the darker background. On the otherhand, at lower consantrations of eosin less than 0.1 %, despate no flame cells motility and protoscolex contractility, dead protoscolex would be misidentified as “ alive”. CONCLUSION: Eozin is a dye which is cheap, less toxic, easily prepared and available at anywhere. Because of those properties, it is prefered for the viability test of protoscolex and is recommended to use 0.1 % and 1 % concentrations for the ideal staining with helping to viability determination.
