Amaç: Bu çalışmanın amacı deneysel olarak siklosporin A (CsA) uygulanan ve diş çekimi yapılan sıçanlarda sistemik resveratrol (RSVL) uygulamasının çekim soket iyileşmesi üzerine etkilerini histopatolojik ve immünohistokimyasal olarak incelemektir. Materyal ve metot: Çalışmada 72 adet Sprague Dawley cinsi dişi sıçan kullanıldı. Denekler 18 sıçandan oluşan 4 ana gruba ayrıldı: Kontrol, CsA, RSVL, CsA+RSVL grubu. CsA ve CsA+RSVL gruplarındaki sıçanlara CsA subkutan enjeksiyonla günlük 10 mg/kg dozunda uygulandı. 8'inci gün, tüm sıçanların alt sol 1. ve 2. büyük azı dişleri çekildikten sonra her ana grup 9'ar denekten oluşan 2 alt gruba ayrıldı. RSVL enjeksiyonu, deney gruplarındaki sıçanların diş çekimleri yapıldığı gün intraperitoneal yolla (10 µmol/kg) uygulanmaya başlandı. Her gruptan 9 denek 14. günde kalanı 28. günde sakrifiye edildi. Deneklerin sol mandibulaları çıkartılıp histopatolojik incelemeye alındı. Elde edilen veriler istatistiksel olarak değerlendirildi. Bulgular: Çalışmamızda elde edilen bulgulara göre gruplar arasında yeni oluşan kemik dokusu açısından 14. ve 28. günlerde istatistiksel olarak anlamlı farklılık gözlendi. RSVL uygulanan grupta bütün diğer gruplara göre daha fazla kemik yapımı görüldü. Buna karşılık CsA uygulanan grupta diğer gruplara göre daha az kemik yapımı saptandı. Ayrıca CsA ve RSVL'ün beraber uygulandığı grupta yeni kemik yapımının CsA grubu ile benzer ve diğer gruplara göre daha az olduğu belirlendi. İmmünohistokimyasal analizde osteokalsin ve osteopontin düzeylerinin 14. ve 28. günlerde RSVL grubunda diğer gruplara göre daha yüksek olduğu görülmüştür. Ayrıca CsA+RSVL grubunun 28. gün osteokalsin ve osteopontin düzeyleri CsA grubundan anlamlı düzeyde yüksek bulunmuştur. Sonuçlar: CsA uygulamasının diş çekim soket iyileşmesi üzerine olumsuz etkilere sahip olduğu saptanırken RSVL uygulamasının soket iyileşmesini stimüle ettiği tespit edilmiştir. Bu çalışmada CsA ve RSVL'nin aynı anda uygulanmasıyla yeni kemik yapımında artış olduğu ancak bu artışın istatistiksel olarak anlamlı olmadığı gözlenmiştir.
Aim: The aim of this study is to examine histopathologicaly and immunohistochemically the effects of resveratrol (RSVL) on the healing of extraction sockets after molar tooth extractions in cyclosporine A (CsA) administered rats. Material and Method: 72 Spargue Dawley female rats were used in our study. The animals were divided into four main groups respectively: Control, CsA, RSVL, CsA+RSVL. CsA and CsA+RSVL groups received 10 mg/kg/day CsA via subcutaneous injection. Eight days following the injection left mandibular first and second molar teeth were extracted in each group. Following extraction, each of all main group were divided in to 2 subgroups. RSVL in a dosage of 10 µmol/kg were given intraperitoneally for 14 and 28 days. Animals were sacrificed 14th and 28th days postoperatively and left hemimandibles were removed and prepared for histopathological examination. Histopathological findings were statistically analyzed. Results: According to the results of our study, there was diferences in new bone formation between groups on 14th and 28th days, which was statistically significant. In RSVL administered group new bone formation was significantly more than others groups. On the other hand, new bone formation was significantly less in CsA group than others groups. Additionally, in group in which CsA and RSVL were applied together, new bone formation was similar to the CsA group and less than all other groups. Immunohistochemical analysis showed higher levels of osteocalcin and ostoepontin in the RSVL group than in the others groups. In CsA+RSVL group osteoocalcin and osteopontin levels was also significiantly higher than CsA group on 28th days. Conclusion: While CsA administration severely impaired the healing of extraction sockets in rats, RSVL administration stimulated the healing of extraction sockets. When CsA and RSVL were combined together there was increasing new bone regeneration but this was not statistically significant.
