Amaç: NaF'ın diş sağlığı için tedavi edici olarak kullanılması sonucu vücuda alınmasının yanısıra topikal uygulamalar, içme suyuna katılması veya tablet gibi sistemik kullanımları ile de NaF'e maruziyet oluşabilmektedir. Çalışmamızda, NaF maruziyeti sonucu testis dokusunda meydana gelebilecek hasar ve bu hasara karşı quercetinin koruyucu etkinliğini histolojik ve biyokimyasal yöntemlerle incelemeyi amaçladık. Gereç ve Yöntem: Çalışmamızda kontrol grubu, mısır yağı grubu, NaF grubu, quercetin grubu ve NaF+quercetin grubu sıçanların testis dokularında histolojik ve immunohistokimyasal yöntemler, testis dokusu ve serumda ise biyokimyasal yöntemler ile incelemeler yapılmıştır. Bulgular: NaF grubu HE boyamalarda, seminifer tübüllerin sınırlarında düzensizlik, germinatif epitelde dejenerasyon, dökülme, lümende hücresel artefaktlar izlendi, bazal membrandan ayrılma gözlenirken, NaF+Qr grubunda bu bulguların gözlenmediği tespit edildi. NaF+Qr grubunda seminifer tübüller genellikle normal histolojik görünümde olmakla birlikte yer yer düzensiz sınırlı seminifer tübüller ve bazal membranlarında incelme, epitelde dökülme gibi bulgular da izlendi. Kontrol grubuna kıyasla C-3 boyanmada NaF grubunda pozitif boyanmada artış gözlendi. NaF+Qr grubunda ise kontrol grubuna kıyasla immunoreaktivite artışı mevcut olup NaF grubuna kıyaslandığında ise C-3 boyanma daha az olarak izlendi. NaF grubunda kontrol grubundan farklı olan biyokimyasal parametrelerin Naf+QR grubunda kontrol grubuna benzer olduğu tespit edildi. Biyokimyasal olarak oksidatif stres parametreleri NaF grubunda artmış olarak gözlenirken antioksidan sistem parametreleri ve hormon düzeyleri de azalmış olarak izlendi. NaF+QR grubunda ise bu değerlerde düzelme gözlendi. Sonuçlar: Bulgular değerlendirildiğinde, NaF maruziyeti uyguladığımız grupta testis dokusunda histolojik ve biyokimyasal hasarların yanında kan testosteron, LH ve FSH hormon düzeylerinde azalma izlendi. NaF+QR grubu değerlendirildiğinde, quercetin uygulamasının, NaF maruziyetine bağlı testiküler hasarı azaltma ve bozulan hormonal düzeylerin dengelenmesi yönünde etkili olduğu tespit edilmiştir. Objective: NaF is used as a therapeutic agent for dental health, as well as taken to body in many ways. NaF exposure may occur as a result of topical applications, introduction to drinking water or systemic use such as tablets. It has been stated that damage to various organs and tissues may occur as a result of this exposure. In our study, we aimed to investigate the protective effect of quercetin with the antioxidative, free radical scavenging and cytoprotective effects on the testis tissue as a result of NaF exposure and histological and biochemical methods. Materials and Methods: In our study, histological and immunohistochemical methods, and serum biochemical methods were performed in testis tissues of control group, corn oil group, NaF group, quercetin group and NaF + quercetin group rats. Results: Hematoxylin-eosin staining showed irregularity at the boundaries of the seminiferous tubules, degeneration of the germinative epithelium, separation from the basement membrane. Caspase 3 staining showed an increase in the Naf group whereas it was nearly normal in Q+Naf group. Conclusions: When the findings were evaluated, blood testosterone, LH and FSH hormone levels were decreased in addition to histological and biochemical damages in the testis tissue in the NaF exposure group. For this reason, it is thought that the use of quercetin in people with endemic fluorosis due to shaip to source waters containing high fluoride in our country or due to dental treatment may be contributing to the prevention of the decrease of testosterone levels in histological and biochemical aspects of testicular tissue in individuals who have been exposed to NaF exposure.
