N-Heterosiklik karben (NHC) ligantları; güçlü σ verici ve zayıf π alıcı özellikleri, azot atomu üzerindeki sübstitüyentlerin sterik ve elektronik özelliklerinin ayarlanabilirliği, metallerle kararlı bağlar oluşturabilme gibi eşsiz özelliklere sahiptir. Organik ve organometalik kimyanın önemli bir sınıfı olan NHC'ler ve onların metal kompleksleri tıbbi kimya, malzeme kimyası ve kimyasal katalizde yaygın olarak kullanılmaktadır. Florun bir moleküle dâhil edilmesi, bileşiğin konformasyon, pKa, içsel potansiyeli, membran geçirgenliği, metabolik yolları ve farmakokinetik özelliklerini verimli bir şekilde etkileyebilir. Ayrıca, flor gruplarının hedef moleküllere yerleştirilmesi için verimli yöntemlerin geliştirilmeside önemli bir araştırma alanı haline gelmiştir. Bu tez çalışmasının temel amacı, elektronegatifliği en yüksek olan flor atomunun karben öncülü ve onların komplekslerine dâhil edilmesi ile sentezlenecek ilaç öncüllerinin lipofilik özelliğini etkileyerek hem vücutta hücre içi geçirgenliği hem de biyolojik aktivitesini etkileyecek olması ve yeni nesil ilaçların sentezine katkı sunmasıdır. Bu tez kapsamında; 1) Florlu benzimidazolyum tuzları (2a-h), NHC-Ag(I) (3a-g), NHC-Pd(II)-piridin (4a-g), NHC-Pd(II)-(2-aminopiridin) (5a-d), kompleksleri sentezlenmiş ve yapıları uygun spektroskopik yöntemler ile aydınlatılmıştır. 2) Florlu benzimidazolyum tuzlarının (2a-h) enzim inhibisyon aktiviteleri incelenmiştir. 3) Moleküler yerleştirme çalışması yapılmıştır. 4) In siliko ADMET çalışması yapılmıştır. N-heterocyclic carbene (NHC) ligands have unique properties such as strong σ-donor and weak π-acceptor properties, tunability of steric and electronic properties of substituents on the nitrogen atom, and the ability to form stable bonds with metals. As an important class of organic and organometallic chemistry, NHCs and their metal complexes are widely used in medicinal chemistry, materials chemistry, and chemical catalysis. The judicious incorporation of fluorine into a molecule can efficiently influence its conformation, pKa, intrinsic potential, membrane permeability, metabolic pathways, and pharmacokinetic properties. Consequently, the development of efficient methods for the insertion of fluorine groups into target molecules has become an important area of research. The main purpose of this thesis study is that the inclusion of the fluorine atom with the highest electronegativity in the carbene precursor and their complexes will affect the lipophilic properties of the drug precursors to be synthesized, which will affect both the intracellular permeability and biological activity in the body and contribute to the synthesis of new generation drugs. Within the scope of this thesis; 1) Fluorinated benzimidazolium salts (2a-h), NHC-Ag(I) (3a-g), NHC-Pd(II)-pyridine (4a-g), NHC-Pd(II)-(2-aminopyridine) (5a-d) complexes were synthesized and their structures were elucidated by appropriate spectroscopic methods. 2) The inhibition activities of the enzyme of fluorinated benzimidazolium salts (2a-h) were investigated. 3) Molecular docking study has been done. 4) In silico ADMET study was performed.
