Kadmiyum'un Neden Olduğu Karaciğer, Akciğer ve Böbrek Hasarı Üzerine Sildenafil'in Koruyucu Etkisi Amaç: Bu çalışmanın amacı kadmiyum ile indüklenen karaciğer, akciğer ve böbrek toksisitesine karşı sildenafilin koruyucu etkisini incelemektir. Materyal ve Metot: Sprague Dawley cinsi dişi sıçanların (n=28) kadmiyumla oluşturulmuş karaciğer, akciğer ve böbrek doku toksisitesi için serum AST, ALT, kreatinin, BUN ve CK-MB ile oksidatif stres göstergelerinden doku TBARs, GSH, T-SH ve enflamasyonda kötü prognoz göstergesi sayılan doku IL-6, TNF-α düzeyleri ile histopatolojik durum değerlendirilmiştir. Bulgular: Kadmiyum'un TNF-α, IL-6 ve TBARs düzeylerini kontrol grubuna göre istatistiksel açıdan anlamlı bir şekilde artırdığı (p<0.05, p<0.001), GSH ve T-SH düzeylerini ise anlamlı derecede azalttığı tespit edilmiştir (p<0.001). Sildenafil+kadmiyum uygulamasında ise, kadmiyum grubuna göre TBARs düzeyi anlamlı derecede azalırken (p<0.05), GSH ve T-SH düzeyleri anlamlı derecede artmıştır (p<0.001). TNF-α ve IL-6 düzeyleri değerlendirildiğinde, sildenafil+kadmiyum grubunda, kadmiyum uygulanan gruba göre anlamlı derecede düştüğü gözlenmiştir (p<0.001, p<0.01). Histopatoloji değerlendirmesinde ise sildenafil uygulaması, kadmiyumun neden olduğu doku harabiyetini azaltmıştır. Sonuç: Sildenafil'in, kadmiyum'un neden olduğu çoklu doku hasarına karşı koruyucu etkisinin antienflamatuar özelliğinden kaynaklandığı söylenebilir. Protective Effect of Sildenafil on Cadmium-caused Liver, Lung and Kidney Injury Aim: The aim of this study was to investigate the protective effect of sildenafil against cadmium-induced liver, lung and kidney toxicity. Material and Method: Sprague Dawley female rats (n=28) are for cadmium-induced liver, lung and kidney tissue toxicity, biochemical parameters serum AST, ALT, creatinine, BUN and CK-MB, tissue TBARs from lipid peroxidation indicators, tissue GSH and T-SH levels from tissue antioxidant parameters, tissue IL-6 and TNF-α levels which are considered as indicators of poor prognosis in inflammation and histopathological status were evaluated. Results: It was found that cadmium significantly increased TNF-α, IL-6 and TBARs levels (p<0.05, p<0.001), GSH and T-SH levels were significantly decreased compared to the control group (p<0.001). In Sildenafil + cadmium application, TBARs decreased significantly compared to cadmium group (p<0.05), GSH and T-SH levels were significantly increased (p<0.001). When TNF-α and IL-6 levels were evaluated, it was observed that sildenafil+cadmium group was significantly decreased compared to cadmium group (p<0.001, p<0.01). In the histopathological evaluation, sildenafil application decreased the cadmium-induced tissue damage. Conclusion: The protective effect of sidenafil against multiple tissue damage caused by cadmium can be said to be due to its anti-inflammatory properties.
