Amaç: MOTS-c 2015 yılında keşfedilmiş, 16 aminoasitli peptit yapılı bir hormondur. Yapılan çalışmalarla MOTS-c'nin obezite ve insülin direncini azalttığı ve hücre metabolizması arttırdığı rapor edilmiştir. Bu çalışma sıçanlara icv MOTS-c infüzyonunun HHT aksı ve enerji kullanımı üzerindeki etkilerini araştırmak amacıyla yapıldı. Materyal ve Metot: 40 adet Wistar-Albino cinsi erkek sıçan 4 gruba ayrıldı (n=10). Kontrol grubu dışındaki sıçanların, lateral ventrikülerine 14 gün süresince 5µl/saat (Sham grubuna yBOS, uygulama gruplarına 10 ve 100 µM MOTS-c) infüzyon gerçekleştirildi. Deney süresince sıçanların günlük vücut ağırlığı ve yem tüketimi ölçüldü. Deney sonunda sıçanlar dekapite edilerek hipotalamus, kas, kan ile beyaz ve kahverengi yağ doku örnekleri toplandı. Hipotalamus dokusunda TRH, beyaz ve kahverengi yağ dokuda UCP1, kas (biceps) dokuda ise UCP3 mRNA seviyesi RT-PZR yöntemi ile belirlendi. Western blot analiz yöntemi ile de beyaz ve kahverengi yağ dokuda UCP1, kas dokuda ise UCP3 protein düzeyi belirlendi. Alınan kan örneklerinden ELISA yöntemi kullanılarak serum TSH, T3 ve T4 hormon seviyeleri belirlendi. Bulgular: MOTS-c infüzyonunun sıçanlarda yem tüketimini arttırdığı (p<0.05) vücut ağırlığında ise değişikliğe neden olmadığı belirlendi. MOTS-c infüzyonu sıçanların TRH mRNA düzeyinde değişikliğe neden olmazken, serum TSH, T3 ve T4 hormon seviyesini azalttığı görüldü (p<0.05). Öte yandan MOTS-c'nin sıçanların beyaz ve kahverengi yağ dokularında UCP1, kas dokusunda ise UCP3 mRNA ve protein düzeyini arttırdığı tespit edildi (p<0.05). Sonuç: Bu çalışmanın sonuçları MOTS-c'nin HHT aksında görev alan dokular ile kas ve yağ dokusunda (beyaz ve kahverengi) fizyolojik roller üstlendiğini göstermektedir. Özellikle HHT aksında baskılayıcı tarzda ortaya çıkan etki ile periferal enerji kullanımındaki aktive edici etkilerini hangi fizyolojik yolaklar üzerinden gösterdiği halen gizemini korumaktadır. Effects of Intracerebroventricular MOTS-c Infusion on Hypothalamus-Pituitary-Thyroid Axis and Energy Use Aim: MOTS-c was discovered in 2015 and is a peptide hormone with 16 amino acids. Studies have shown that MOTS-c reduces obesity and insulin resistance and increases cell metabolism. This study was conducted to investigate the effects of icv MOTS-c infusion on HHT axis and energy use in rats. Material and Method: Forty male Wistar-Albino rats were divided into 4 groups (n=10). The rats outside the control group were infused into the lateral ventricles for 5 days/hour (yBOS in the Sham group, 10 and 100 µM MOTS-c in the sham group) for 14 days. During the experiment, daily body weight and food consumption of rats were measured. At the end of the experiment, rats were decapitated and hypothalamus, muscle, blood and white and brown fat tissue samples were collected. TRH in hypothalamus tissue, UCP1 in white and brown fat tissue and UCP3 mRNA in muscle (biceps) tissue were determined by RT-PCR method. UCP1 protein level was determined in white and brown fat tissue and UCP3 protein level in muscle tissue by Western blot analysis method. Serum TSH, T3 and T4 hormone levels were determined by ELISA method. Results: It was determined that MOTS-c infusion increased food intake in rats (p <0.05) and did not cause any change in body weight. While MOTS-c infusion did not cause any change in TRH mRNA level, it was seen that it decreased serum TSH, T3 and T4 hormone levels (p <0.05). On the other hand, it was found that MOTS-c increased UCP1 and protein UCP3 mRNA and protein levels in white and brown fat tissues of rats (p <0.05). Conclusion: The results of this study show that the MOTS-c plays a physiological role in the tissues involved in the HPT axis and muscle and adipose tissue (white and brown). In particular, the suppressive effect on the HPT axis and the physiological pathways it demonstrates its activating effects in the use of peripheral energy remain a mystery.
