    Benzimidazole derivatives as a new scaffold of anticancer agents: Synthesis, optical properties, crystal structure and DFT calculations

    The absolute necessity to fight some class of tumour is perceived as serious health concerns, and the discovery and development of effective anticancer agents are urgently needed. So, the novel benzimidazole derivatives (2a-b) were designed, synthesized, with their structures rigorously characterized using single X-ray crystallography, FT-IR, UV, and NMR spectroscopy, alongside elemental analysis. The geometric structures were optimized using density functional theory (DFT) calculations performed at the ?B97X-D/cc-pVDZ level, yielding good agreement with experimental XRD data. The studied salt complexes exhibited the ability to absorb UV light at 275 nm. Furthermore, anticancer activity of the compounds was screened against (MDA-MB-231, MCF-7, HT-29 and healthy cell line (HF)) and revealed the remarkable efficacy of select newly synthesized Benzimidazole derivatives (2a-b). Compound 2a showed relative significant higher cytotoxicity (165.02) in MDA-MB-231 cancer cell line. This underscores their promising potential in therapeutic applications, affirming their role as valuable contenders in the pursuit of novel anticancer agents. © 2024 The Authors

    Evaluation of the Appropriateness of Antimicrobial Drug Dosages in Intensive Care Unit Patients

    The purpose of this study was to investigate the factors that required dose adjustments of antimicrobial drugs in intensive care unit (ICU) patients and to identify the drugs that required the most dose adjustments. The current prospective study was conducted in the reanimation ICU with 26-bed capacity of a university-af-filiated hospital from September to December 2022. Two clinical pharmacists on duty examined patients’ antimicrobial drug dosages daily. The acceptance status of the recommendations and the patients’ demographic information were recorded. The study involved 133 ICU patients, and antimicrobial drug recommendations were made for 48 patients, 31 (64.6%) of whom were male. The median (IQR) age of the 48 patients was 67 (54–77). The count of recommendations was 94, and the acceptance rate was 100%. The recommendation rates were as follows: 71.3% for renal function, 11.7% for presence of continuous renal replacement therapy, 10.6% for indication, 4.3% for body weight, and 2.1% for loading dose. The top 3 drugs for which recommendations were made the most were colistin (21.3%), meropenem (18.1%), and piperacillin-tazobactam (12.8%). The most troublesome drug was colistin, which is frequently used to treat Acinetobacter pneumonia. Clinical pharmacist and physician collaboration may help rationalize ICU antimicrobial drug use. © 2024, Hacettepe University, Faculty of Pharmacy. All rights reserved.

    Synthesis, electrochemical, biological and catalytical studies of half-sandwich Ru(II)-NSHC complexes bearing benzothiazol-2-ylidene

    The half-sandwich ruthenium(II)-NSHC complexes (2a-l) bearing benzothiazol-2-ylidene were synthesized by in situ deprotonation of N-substituted benzothiazolium salts (1a-l) with Ag2O and of [(p-cymene)Cl2Ru]2, in dichloromethane under mild conditions. However, 1m and 1n under the same conditions gave N-coordinated Ru(II) complex (3). The structures of the compounds were elucidated by NMR, FT-IR, UV–Vis, HR-MS spectroscopic methods and elemental analysis. In addition, the structures of 2k and 3 were studied by X-ray crystallography. The electrochemical properties of the complexes were investigated. Ru(II)-NSHC complexes have been used as catalysts in the transfer hydrogenation of carbonyls to secondary alcohols in the presence i-PrOH/KOH. Additionally, the effects of structural differences arising from different alkyl groups on the nitrogen atom in the benzothiazole skeleton in Ru(II)-NSHC complexes on anticancer, antifungal and antimicrobial activity were also examined. © 2024 Elsevier B.V.
