Elde edilen parametreler, uluslararası yayınlarda sunulmuş benzer bilgisayar sistemleri ile elde edilmiş verilerle karşılaştırmaları yapılmıştır. Çalışmada aynalı sazan (Cyprinus carpio) türü balıklara ait sperm örnekleri incelenmiştir. İncelenen sperm hücrelerine ait hareketlilik parametreleri sırasıyla VSL: 74.05 ?m/sn, VCL: 115.18 ?m/sn, VAP: 63.85?m/sn, BCF: 10.75 Hz ve ALH: 19.28?m olarak hesaplanmıştır. Sonuç olarak, İmaj J-Fiji programı ile balıklarda sperm hücresi yakalama, işleme ve değerlendirme işlemlerinin kolay, uygulanabilir olduğu ve balık üretim merkezlerinde erkek damızlık balıkların sperm kalitesinin belirlenmesinde pratik ve hızlı bir yöntem sunduğu söylenebilir.Öğe Investigation of Toxicity Properties of Flower-like ZnO Nanoparticles on Cyprinus carpio Sperm Cells Using Computer-Assisted Sperm Analysis (CASA)(2018) Özgür, Mustafa Erkan; Ulu, Ahmet; Balcıoğlu, Sevgi; Özcan, İmren; Okumuş, Fatih; Köytepe, Süleyman; Ateş, BurhanAbstract: This study was carried out to clarify the toxicity properties of Flower-like zinc oxide nanoparticles (ZnO NPs) on collected sperm samples from Cyprinus carpio using computer-assisted sperm analysis (CASA). Flower-like ZnO NPs were successfully synthesized using a simple system at 120?C for 5 h by the microwave-assisted solvothermal techniques. The synthesized ZnO NPs were characterized for its structural, morphological and thermal properties. After that, the different concentrations of synthesized ZnO NPs (0.001, 0.01, 0.1, 0.5, 1 ppm) were examined with Cyprinus carpio sperm samples collected from Karakaya reservoir. The spermatozoa motility parameters which contain to curvilinear velocity (VCL), straight line velocity (VSL), average path velocity (VAP), straightness (STR), amplitude of lateral head displacement (ALH), and beat cross frequency (BCF) was computed and recorded by CASA. According to the results, while VCL, VAP and BCF were statistically significant (P<0.05), VSL, ALH and STR were not statistically significant (P>0.05). In addition, the EC50 values of VCL and VAP parameters were calculated 0.56 ppm and 0.004 ppm, respectively.Öğe A Novel Computer Assisted Sperm Analyzer for Assessment of Spermatozoa Motility in Fish; BASA-Sperm Aqua(2019) Özgür, Mustafa Erkan; Okumuş, Fatih; Kocamaz, Adnan FatihAbstract: This study was conducted to determine the working principle and operability of the BASA-SpermAqua module software of the newly developed computer-assisted sperm analysis system (BASA) for theevaluation of spermatozoa motility in fish. Semen samples of trout (Oncorhynchus mykiss) species wereexamined for this purpose. Sperm motility parameters such as VSL (?m/s), VCL (?m/s), VAP (?m/s), LIN (%),BCF (Hz), ALH (?m) and MAD (o) were examined. The investigated parameters were compared with data whichanalyzed in similar computer systems and published in international manuscripts. Finally, the BASA-SpermAqua has been found to be a software that performs its functions very quickly and practical and producesaccurate and understandable results in the determining sperm quality parameters of fish.
