    Antidepressant effect of Gentiana olivieri Griseb. in male rats exposed to chronic mild stress

    Background: The flowering parts of Gentiana olivieri, known as 'Afat' in the southeastern Anatolia region of Turkey, are used as a tonic, an appetizer, and for the treatment of several mental disorders, including depression. The purpose of this study is to investigate the antidepressant effect of G. olivieri ethanol extract (GOEE) in a chronic mild stress-induced rat model, which was used to mimic a depressive state in humans, and to compare the effect with that of imipramine. Methods: Male Sprague-Dawley rats were randomly divided into six groups: control, stress, treated with imipramine (positive control) and treated with GOEE at three different (200, 500, 1000 mg/kg) doses groups. The rats in all groups, except the control group, were exposed to chronic mild stress. At the end of the 3-week experimental period, biochemical and behavioral parameters were examined. Results: The results showed that treatment with GOEE or imipramine significantly improved rats' sucrose consumption which was diminished by chronic mild stress, restored serum levels of corticosterone and proinflammatory cytokines (interleukin-6 (IL-6), tumor necrosis factor-alpha (TNF-alpha)), prevented the increase of liver index of rats. Moreover, in the hippocampus tissue, decreased serotonin and noradrenaline levels were significantly increased by treatment with GOEE or imipramine, and antioxidant parameters (thiobarbituric acid reactive substances (TBARS), superoxide dismutase (SOD), and glutathione (GSH)) were significantly improved by treatment with GOEE though not with imipramine. Conclusion: The data demonstrate that G. olivieri may exert its antidepressant activity by improving monoaminergic system disorders, and by favorably affecting the antioxidant, inflammatory and the endocrine mechanisms.

    Antioxidant Effect of Sildenafil on Cadmium-Induced Liver, Lung and Kidney Injury

    The aim of this study is to investigate the protective effect of sildenafilagainst cadmium-induced liver, lung and kidney toxicity. In total of28 Sprague Dawley female rats included in this study were dividedinto four groups: control, Sil, Cd, Sil+Cd groups. CdCl2 and Silcitrate were dissolved in distilled water, the injection volume adjustedto 0,5 ml / rat. On the 7th day, a single dose of 3,7 mg/kg Cd wasinjected ip in the morning and Sil was administered in drinkingwater for 10 days. On the 10th day of the experiment, 72 hours afterCd administration, the rats were sacrificed under anesthesia and thelungs, kidneys and livers of the rats were isolated and Thiobarbituricacid reactive substances (TBARs), GSH, total thiol (T-SH) levels inthese tissues were evaluated. Tissue damage was assessed by serumalanine aminotransferase (ALT), aspartate aminotransferase (AST),creatinine, blood urea nitrogen (BUN) and creatine kinase (CK-MB). It was found that cadmium significantly increased AST, ALT,creatinine, BUN, CK-MB and TBARs levels compared to the controlgroup, and significantly decreased GSH and T-SH levels. In sildenafil+ cadmium administration, TBARs levels significantly decreasedwhile GSH and T-SH levels significantly increased compared tocadmium group. The protective effect of sildenafil against multipleorgan damage caused by cadmium may be due to its antioxidantproperties.

    Investigation of antidepressant -like effect of pari-flo product on forced swimming test in balb-c mice

    Abstract: Pari-flo is a commercial preparation in the form of a solution containing standardized Passiflora incarnata plant extract. We hypothesized that Pari-flo may have antidepressant potential with its content. Therefore, the aim of this study was investigation of the antidepressant-like effect of Pari-flo product comparing with imipramine based on Forced Swimming Test (FST) in Balb-c Mice. Forty-two male Balb-c mice were divided into 6 equal groups as follows: group 1 (control) were fed with normal rat chow and tap water, groups (2, 3, 4 and 5) were given 125, 250, 500 and 1000 mg/kg/day dose Pari-flo respectively, group (6) were given 15 mg/kg/day dose imipramine. After 7 days of treatment, the FST was carried out. Pari-flo significantly reduced the immmobility time while increased the climbing time in FST at the dose of 500 and 1000 mg/kg/day. At the 1000 mg/kg/day dose of Pari-flo was increased the swimming time in FST. Imipramine was also increased the swimming and climbing time, while reduced the immobility time. Based on FST test results, it is concluded that Pari-flo product has antidepressant-like activity comparable to those of imipramine in experimental animal models.

    YARA İYİLEŞMESİ VE DİYABETİK YARA TEDAVİSİDE KULLANILAN TIBBİ BİTKİLER

    Öz: Diabetes mellitus insülin salgısında düşüş ve/veya insülinin etkisinde azalmaya bağlı ortaya çıkan, yüksek kan şekeri düzeyi ile karakterize metabolik bir hastalık olup çevresel, genetik ve yaşam biçimi gibi değişik faktörlerin katkısıyla meydana gelmektedir. Diyabet kontrol altına alınmadığı taktirde ilerleyici bir seyir izlemekte, buna bağlı olarak da morbidite ve mortalitede artışa yol açarak toplum ve birey açısından büyük sağlık sorunu oluşturmaktadır. Diyabetik nöropati, periferal vasküler hastalıklar ve anormal hücresel aktiviteler diyabetik yara iyileşmesini engelleyen temel faktörlerdir. Son yıllarda dünya genelinde, sağlıklı bir yaşamın sürdürülmesi ve çeşitli hastalıkların tedavisinde tıbbi bitkilerin kullanımı artış göstermektedir. Yara iyileşmesinin gecikmesi ve kronik yaralar diyabetin komplikasyonlarından olup günümüz tıbbının ciddi tedavi problemi ve ekonomik sorunları olarak karşımıza çıkmaktadır. Diyabetik yara tedavisi için mevcut yaklaşımlar ve geliştirilen yeni yöntemler kısmen etkili olmakta, bu nedenle kısa sürede iyileşme sağlayacak başka tedavi yöntemlerinin geliştirilmesi önem arz etmektedir. Bu derlemede yara iyileşmesi hakkında genel bilgiler derlenmiş olup, diyabetik yara tedavisinde kullanılan tıbbi bitkilerle yapılan çalışmalar özetlenerek yeni çalışmalara katkı sağlamak amaçlanmıştır. Başlık (İngilizce): WOUND HEALING AND MEDICINAL PLANTS USED IN THE TREATMENT OF DIABETIC WOUNDS Öz (İngilizce): Diabetes mellitus is a metabolic disease characterised by high blood level, occured due to the low secretion of insulin and/or decrease in insulin effect. It happens with the contribution of different factors such as environment, genetic and life style. When diabetes is not taken under control, it runs a progressive course; therefore, causes a big health problem for the society and individuals, with the increase in the morbidity and mortality. Diabetic neuropathy, periferal vascular diseases and abnormal cellular activities are the main factors which a prevent the healing of diabetic wounds. The use of medicinal plants has recently been increased throughout the world for the maintenance and improvement of health and for the treatment of various human conditions and diseases. Delayed wound healing and chronic wounds are the complications of diabetes mellitus and are the major therapeutic and economic issues in medicine today. Development of different methods that would yield a recovery in a short period of time merits attention by virtue of limited effectiveness of current approaches for treatment of diabetic wounds. In this review, we summarized general knowledge of wound healing and the current treatment approaches to diabetic wounds and previous works related to medicinal plants to contribute the future studies in this field.
