Öğe Fenilboronik asit fonksiyonelli N-heterosiklik karben öncüllerinin sentezi, karakterizasyonu ve biyolojik özelliklerinin incelenmesi(İnönü Üniversitesi, 2022) Kuruçay, AliN-heterosiklik karbenler (NHC), singlet karbenlerden oluşan ve içerisinde azot atomu bulunduran heterosiklik bileşiklerdir. Bu tezde boronik asit fonksiyonel grubu içeren benzimidazolyum tuzlarının sentezi, karakterizasyonu, antikanser, genotoksisite, antibakteriyel ve enzim inhibisyonu özellikleri araştırılmıştır Çalışma kapsamında meta-fenilboronik asit sübstitüentli benzimidazolyum tuzları (1a-f) sentezlenmiştir. Sentezlenen tuzların MCF-7 Hücre Hattı üzerinde 1a-f tuzlarının ve cisplatin ilacı için IC50 µg/mL değerleri sırasıyla 37.40±1.70, 54.17±2.77, 29.57±3.04, 32.27±4.24, 29.90±2,89, 38.97±2.44 ve 82.02±6.19 olarak bulunmuştur. A549 Hücre Hattı üzerinde ise 1a-f tuzlarının ve cisplatin için IC50 µg/mL değerleri sırasıyla 38.83±3.17, 51.17±5.84, 17.14±3.72, 31.86±3.53, 29.24±2.86, 24.18±3.99 ve 91.31±3.65 olarak tespit edilmiştir. HCT116 Hücre Hattı için 1a-f tuzlarının ve cisplatin için IC50 µg/mL değerleri sırasıyla 29.20±2.33, 52.78±2.22, 34.29±1.97, 30.42±4.52, 15.31±3.81, 36.04±1.38 ve 106.57±6.77 olarak bulunmuştur. Genotoksik çalışmalarda pBR322 plazmiti üzerinde artan konsantrasyona rağmen bir etkisi görülmemiştir. B. subtilis ve E. coli bakterilerinde standart antibiyotiklerle karşılaştırılabilir düzey öldürücü etki göstermemiştir. Enzim inhibisyonu testinde 1a-f tuzlarının ve allopurinol için IC50 µg/mL değerleri sırasıyla 1.062±0.038, 1.226±0.021, 0.761±0.024, 0.811±0.033, 0.637±0.011, 0.591±0.016 ve 1.685±0.051 olarak bulunmuş olup ksantin oksidaz enzimine allopurinolden daha fazla inhibisyon etkisinin olabileceği tespit edilmiştir. Asetilkolinesteraz enzimine ise donepezilden daha fazla inhibisyon özelliği göstermiş ve bu değerler 1a-f tuzlarının ve donepezil ilacı için IC50 µg/mL değeri sırasıyla 0.861±0.045, 1.088±0.049, 0.976±0.058, 0.862±0.033, 0.903±0.011, 1.029±0.089 ve 3.172±0.661 olarak bulunmuştur. Sonuç olarak boronik asit fonksiyonelli NHC öncüllerinin biyolojik olarak aktif bileşikler olduğu ve özellikle enzim inhibisyonları yolu ile hücreler üzerinde biyolojik olarak aktivite sergiledikleri tespit edilmiştir.Öğe THE PREPARATION OF VARIOUS TEAR-CONTAINING CREAMS AND INVESTIGATION OF THEIR ANTIBACTERIAL PROPERTIES(2021) Gürses, Canbolat; Uyan, Melek; Kuruçay, Ali; Paşahan, AzizAbstract: In our research, the antibacterial properties of tear which protect our eyes from many infections have been investigated by using Bacillus subtilis, Pseudomonas aeruginosa, Staphylococcus haemolyticus and Staphylococcus epidermidis bacteria. In addition, the antibacterial effect of creams in different formulations containing vaseline, also known as petroleum jelly, prepared by mixing olive oil and lemon juice which have been known and used throughout history with tears have been investigated. The antibacterial effect has been investigated measuring the zone of inhibition diameters, also known as the Kirby-Bauer method. It has been determined that all samples showed antibacterial effects except Staphylococcus epidermidis to which only tear was applied. The highest antibacterial activity with tear containing cream (vaseline and olive oil) has been observed for Pseudomonas aeruginosa and Staphylococcus epidermidis bacteria. The basic physicochemical properties of these structures, their structural characterization, thermal properties and morphological properties were investigated. Toxicity properties of constructs synthesized on zebrafish (Danio rerio (Hamilton)) offspring were determined in vivo. In addition, in vitro toxicity tests were performed on L929 fibroblast cells. When the general properties of these structures were examined. Structural and thermal properties were better with increasing cross-linker rates ratios (1%, 3%, 5%). According to the toxicity test performed on zebrafish juveniles; GA-PEG-Epox (1%) constructs are non-toxic to zebrafish juveniles. The mortality rate of GA-PEG-Epox (3%) and GA-PEG-Epox (5%) structures was observed as 12.5% and 20.8%, respectively. It was observed that the structures were not toxic to zebrafish juveniles. MTT test performed on L929 fibroblast cells, high cell viability (>90%) was observed in all synthesized structures. These results are evaluated as Grade 1 according to ISO standards.
