    Assessment of Association Rule Mining Using Interest Measures on the Gene Data

    Aim: Data mining is the discovery process of beneficial information, not revealed from large-scale data beforehand. One of the fields in which data mining is widely used is health. With data mining, the diagnosis and treatment of the disease and the risk factors affecting the disease can be determined quickly. Association rules are one of the data mining techniques. The aim of this study is to determine patient profiles by obtaining strong association rules with the apriori algorithm, which is one of the association rule algorithms. Material and Method: The data set used in the study consists of 205 acute myocardial infarction (AMI) patients. The patients have also carried the genotype of the FNDC5 (rs3480, rs726344, rs16835198) polymorphisms. Support and confidence measures are used to evaluate the rules obtained in the Apriori algorithm. The rules obtained by these measures are correct but not strong. Therefore, interest measures are used, besides two basic measures, with the aim of obtaining stronger rules. In this study For reaching stronger rules, interest measures lift, conviction, certainty factor, cosine, phi and mutual information are applied. Results: In this study, 108 rules were obtained. The proposed interest measures were implemented to reach stronger rules and as a result 29 of the rules were qualified as strong. Conclusion: As a result, stronger rules have been obtained with the use of interest measures in the clinical decision making process. Thanks to the strong rules obtained, it will facilitate the patient profile determination and clinical decision-making process of AMI patients.

    Effect of humanine on the Notch signaling pathway in myocardial infarction

    Background/aim: By applying humanin (HN) before myocardial infarction (MI), its protection in myocardial injury and the possible roles of its cellular mechanism in the Notch pathway were investigated. Materials and methods: The study was carried out at Firat University Experimental Research Center (12/24/2018-12/23/2019). Spraque-Dawley rats were divided into 10 groups: I (control) (n = 6), II (HN 6 h) (n = 6), III (HN 24 h) (n = 6), IV (HN day 7) (n = 6), V (MI 6 h) (n = 7), VI (MI 24 h) (n = 7), VII (MI day 7) (n = 7), VIII (MI+HN 6 h) (n = 7), IX (MI+HN 24 h) (n = 7), and X (MI+HN day 7) (n = 7). To create MI, 200 mg/kg of isoproterenol (ISO) was administered to the rats subcutaneously. Moreover, 252 mu g/kg of HN was given intraperitoneally (ip) to the rats on its own and before MI. Molecular parameters Notch1, Notch2, Hes1, Hes2, Jagged1, Jagged2, DLL1, and DLL4 were examined using polymerase chain reaction in the heart tissue, Notch1, Hes1, and DLL4 were examined using western blot, while heart tissue was taken for histochemical examinations. Results: The mRNA expression levels of the Notch signaling members (Notch1, Notch2, Hes1, Hes2, Jagged1, Jagged2, DLL1, and DLL4) tended to decrease after MI. The Notch signaling members increased more significantly, especially toward day 7 after HN application before MI. In the western blot anylyses, the Notch1, Hes1, and DLL4 protein levels increased significantly toward day 7 in the groups given HN before MI. Moreover, the serum AST, LDH, CK-MB, and troponin I levels tended to decrease with the application of HN before MI and there was a significant decrease in edema, hemorrhage, and mononuclear cells in the heart tissue at 24 h post-MI and fibrosis on day 7 post-MI. Conclusion: HN administration before MI has a cardioprotective effect on rats via the Notch signaling pathway.

    The inhibition of Src kinase suppresses the production of matrix metalloproteinases in from synovial fibroblasts and inhibits MAPK and STATs pathways

    Abstract: Background/aim: The purpose of this study was to investigate the antiarthritic potentials of the inhibition of Src kinase in vivo and in vitro settings. Materials and methods: Arthritis was induced by intradermal injection of chicken type II collagen combined with incomplete Freund’s adjuvant (collagen induced arthritis [CIA] model) in Wistar albino rats. One day after the onset of arthritis, dasatinib, a potent Src kinase inhibitor, (5 mg/kg/day) was given via oral gavage. Tissue Src, Fyn, MAPK and STAT mRNA expressions were determined by real-time polymerase chain reaction. On the other hand, fibroblast like synoviocytes (FLSs) were harvested patients with rheumatoid arthritis (RA) undergoing surgical knee joint replacement. FLSs were stimulated with cytokines and dasatinib was added in different concentrations. MMP –1, –3, and –13 levels in FLSs culture were determined by ELISA. Results: The tissue mRNA expressions of Src, Fyn, MAPK and STATs were increased in the arthritis CIA group compared to the control group. Their mRNA expressions in the CIA + dasatinib group were decreased and similar in the control group. In in vitro setting, MMP –1, –3, and –13 expressions from FLSs induced by IL-1? and TNF-? were increased, while dasatinib suppressed their productions from FLSs. Conclusion: The present study shows that the inhibition of Src kinase has antiarthritic potentials in both in vivo and in vitro settings. Src kinase inhibition may be candidate to further research in human RA. Key words: Rheumatoid arthritis, collagen induced arthritis, src kinase, matrix metalloproteinase
