Öğe Subkronik formaldehit inhalasyonunun sıçanlarda bazı böbrek enzim aktivitelerine etkisi(Van Tıp Dergisi, 2002) Yılmaz, H. Ramazan; Özen, O. Aslan; Songur, Ahmet; Söğüt, Sadık; Özyurt, Hüseyin; Sarsılmaz, MustafaÖz: Bu çalışmada, subkronik formaldehit inhalasyanunun, sıçan böbreğinde karbohidrat metabolizmasının önemli enzimleri olan Glukoz-6-fosfat dehidrogenaz (G6PD), 6-fosfoglukonat dehidrogenaz (6PGD), Laktat dehidrogenaz (LDH), Malat dehidrogenaz (MDH) ve Heksokinaz (HK) enzim aktivitelerine etkisi araştırıldı. Bu amaçla, 24 adet yetişkin erkek Wistar-Albino sıçan kullanıldı. Bu sıçanların formaldehit inhale ettirilmeyen 8 tanesinden kontrol grubu oluşturuldu. Subkronik formaldehit inhalasyonunun toksik etkisinin araştırıldığı hayvanların 8 tanesine 10 ppm dozunda, diğer 8 tanesine de 20 ppm dozunda 13 hafta süreyle formaldehit inhajle ettirildi. Böbrek dokusundaki enzimlerin aktiviteleri spektrofotometrik olarak tayin edildi. 10 ppm dozu inhale ettirilen sıçanlarda, böbrek 6PGD (p<0.01), LDH (p<0.001) ve MDH (p<0.01) enzim aktivitelerinde kontrole göre önemli bir artma gözlendi. 20 ppm dozu inhale ettirilen sıçanlarda, böbrek HK ve G6PD enzim aktivitelerinde önemli bir azalma bulundu (p<0.05). 6PGD (p<0.05), LDH (p<0.01) ve MDH (p<0-05) enzim aktivitelerinde ise, önemli bir artma gözlendi. Sonuç olarak, erkek sıçanlara inhale ettirilen iki ayrı dozda formaldehitin, böbrek G6PD, 6PGD, LDH, MDH ve HK enzfm aktİvUelerini değiştirerek glukozun esas olarak girebileceği ana metabolik yolları etkilediği söylenebilir. Başlık (İngilizce): The toxic effects of subchronic (13 Weeks) formaldehyde inhalasion on some enzymes in the kidney of male rats Öz (İngilizce): In this study, the toxic effects of subchronic formaldehyde inhalation on the activities of hexokinase (HK), glucose-6-phosphate dehydrogenase (G6PD), 6-phosphogluconate dehydrogenase (6PGH), lactate dehydrogenase (LDH), and malate dehydrogenase (MDH) of the kidney of male rats were investigated. The aim of this study was to evaluate whether enzyme activities are changed in the kidney of rats exposed to subchronic formaldehyde inhalation (13-weeks). Twenty-four male Wistar albino rats were randomly divided into three separate groups. The first eight rats were used as control group. Rats were exposed to atmosphere containing 10 (n=8) and 20 ppm (n=8) formaldehyde for 13 weeks, respectively in group I and group II Spectrophotometric method was used to determine the activities of the enzymes. The results showed a remarkable increase in 6PGD (p<0.01), LDH (p<0.001), and MDH (p<0.01) activities in the kidney tissues from rats exposed to 10 ppm formaldehyde for 13-weeks compared to controls. It was shown significantly that the HK and G6PD activities were reduced (p<0.05) and 6PGD (p<0.01) and MDH (p<0.05) activities were increased in the kidney tissues from rats exposed to 20 ppm formaldehyde for 13 weeks.As a result, it can be suggested that formaldehyde at two different doses inhaled by male rats may affect the main metabolic roots of glucose leading to the changes of the enzyme activities of kidney G6PD, 6PGD, LDH, MDH, and HK. Although underlying mechanism that affects enzyme activities is not clear, it seems more likely to change the activities by direct mechanism on enzyme activity rather than indirect mechanisms such as transcription and translation processes of the enzymes. 2 ATIF 11 Atıf Sayısı ui-button
