Apelin and APJ have also been identi- fied in the testis, hypothalamic nuclei such as arcuate, supraoptic and paraventricular nuclei, implicating roles in the control of reproduction. Therefore, the present study was designed to investigate the effects of chronic central infusion of apelin-13 on LH, FSH and testosterone levels and testis morphology. 21 Wistar–Albino male rats received continuous intracerebroventricular infusion via Alzet osmotic mini pumps filled artificial cerebrospinal fluid (vehicle) or apelin-13 at concentrations of 1 or 10 nmol (10 l/h) for seven days. At the last 90 min of the infusion period, the blood samples were collected at 15 min intervals (0–90 min) for LH and FSH analyses. At the last sampling point, the blood samples were analyzed for testosterone levels. Infusion of high dose apelin-13 significantly suppressed LH release compared with the vehicle values at 30, 60 and 75 min (p < 0.05). However, FSH levels did not significantly differ among the groups. Serum testosterone levels in high dose apelin-13 group were statistically lower than the control group (p < 0.05). In addition, histological examination showed that infusion of high dose apelin-13 significantly decreased the number of Leydig cells compared with the control and lower dose apelin-13 groups (p < 0.05, p < 0.01). Our results suggest that apelin-13 may play a role in the central regulation and decreases testosterone release by suppressing LH secretion. All rights reserved.Öğe Neonatal LPS uygulaması ve üremenin uzun vadeli programlanması: NOS ve Kaspaz-1 inhibisyonlarının rolü(İnönü Üniversitesi, 2012) Tapan, TubaBu tez çalışmasında, neonatal dönemde bakteriyel endotoksin lipopolisakkarit (LPS) uygulanan sıçanlarda üremenin uzun vadeli programlanması incelenmiştir. Ayrıca, LPS'nin etkisine aracılık eden nitrik oksit (NOS) inhibitörü (L-NAME) ve kaspaz-1 inhibitörü (Q-Vd-OPh), neonatal dönemde LPS ile birlikte kullanılarak bu aracıların rollerinin ortaya konması hedeflenmiştir. Bu amaçla, postnatal 7. günde dişi sıçan yavrularına salin, LPS (50 µg/kg), LPS (50 µg/kg)+ L-NAME (40 mg/kg) veya LPS (50 µg/kg)+ Q-Vd-OPh (1 mg/kg) enjeksiyonu yapılmış; prepubertal dönemde (postnatal 30. gün) de salin veya ikinci LPS enjeksiyonu (50 µg/kg) yapılarak oluşturulan stresin puberteye erişme (vajinal açılma), östrus sikluslarının düzeni (simir testi), sitokin üretimi (IL-1ß, TNF-?), ovaryumda foliküler gelişme ve E. coli'ye spesifik anti-LPS antikorlarının üretimi üzerine etkileri belirlenmiştir. Ortalama olarak postnatal 75. günde proöstrus fazında eter anestezisi yapılan sıçanların sağ ovaryumları ve kan numuneleri alınarak deney sonlandırılmıştır. Puberte, iki defa LPS uygulanan grupta gecikmiş fakat NOS ve interlökin (IL)-1ß inhibitörleri pubertenin gecikmesini engellemiştir. LPS enjeksiyonları primordiyal folikül rezervini azaltmış, NOS inhibitörü bu azalmayı önlerken, IL-1ß inhibitörünün bu azalmaya karşı koruyucu bir rolü belirlenememiştir. İki defa LPS enjeksiyonu yapılan tüm gruplarda LPS'ye karşı spesifik antikor yanıtı gözlenmiştir. Serum NO konsantrasyonları tüm gruplarda benzer bulunmuştur. Serum IL-1ß, tümör nekrozis faktör- alfa (TNF-?) konsantrasyonları ise tespit edilebilir düzeyin altında bulunmuştur.
