Öğe Sulfonated N-heterocyclic carbine-silver (I) complexes: Synthesis, characterisation and biological evaluation(Wiley, 2018) Yasar, Seyma; Koprulu, Tugba Kul; Tekin, Saban; Yasar, SedatThe synthesis, characterisation and biological activity of water-soluble Ag(I)-NHC complexes, general formula Na[(NHC)AgCl] where NHC is a sulfonated and sterically hindered N-heterocyclic carbene, is reported. The Ag-NHC complexes (2a-e) were synthesised by reacting the corresponding sulfonated NHC ligands with Ag2O in the presence of NaCl or NaBr in methanol/water (1:1) solution. Synthesised silver (I)-N-heterocyclic carbene complexes have been characterised by NMR, micro-analysis and HRMS spectroscopic methods. The IC50 values of these complexes were determined by a proliferation BrdU enzyme-linked immunosorbent assay (ELISA) against HeLa (human cervix carcinoma), HT29 (human adenocarcinoma) and L929 (mouse fibroblast) cell lines. These complexes have been highlighted as promising and original platforms for building new types of metalodrug. All new water-soluble Ag(I) complexes demonstrated remarkable cytotoxic activity against HeLa, HT29 and L929 cell lines.Öğe Synthesis and investigation of antiproliferative activity of Ru-NHC complexes against C6 and HeLa cancer cells(Tubitak Scientific & Technological Research Council Turkey, 2022) Pasahan, Ramazan; Akkoc, Mitat; Yasar, Seyma; Kul Koprulu, Tugba; Tekin, Saban; Yasar, Sedat; Ozdemir, IsmailThe 2-methylpyridine, 2-diethylaminoethyl, and isopentyl linked a series of symmetric and unsymmetric benzimidazolium salts 2a-e were prepared and used in the synthesis of silver-N-heterocyclic carbene (NHC) complexes (3a-e). The Ru(II)-NHC complexes (4a-h) were synthesized via transmetalation reaction from 3a-e. 4a-h complexes were converted to Ru(II)-NHC. HCl complexes (5a-h) by HCl solution of diethyl ether and characterized by different spectroscopic techniques such as H-1 and C-13 NMR, LC/MS-Q-TOF, FT-IR, elemental analysis, and melting point detection. We examined the effect of the structural difference of complexes on anticancer activity via different arenes and metal centers. Antiproliferative activity of 5a-h and 3a was tested against human cervix adenocarcinoma (HeLa) and rat glioblastoma (C6) cell lines by ELISA assay. The IC50 value of 5b, 5c and 5e complexes exhibited good cytotoxic activity than cisplatin on C6 (14.2 +/- 0.5 mM; 16.2 +/- 0.4 mM; 24.2 +/- 0.7 mM, respectively) and HeLa (11.1 +/- 0.5 mM; 13.7 +/- 0.3 mM; 22.8 +/- 0.8 mM, respectively) cell lines.Öğe Synthesis, characterisation and cytotoxic properties of N-heterocyclic carbene silver(I) complexes(Elsevier Science Sa, 2018) Yasar, Seyma; Koprulu, Tugba Kul; Tekin, Saban; Yasar, SedatA benzimidazolium salt bearing hydroxyethyl group, 1a and a new series of zwitterionic sulphonated benzimidazolium salts, 1b-e were reacted with Ag2O to produce Ag(I)-N-heterocyclic carbene (NHC) complexes, 2a-e respectively. Synthesised silver(I)-N-heterocyclic carbene complexes were fully characterised by H-1 and C-13 NMR, elemental analysis and HRMS spectroscopic methods. Anti-cancer potential of both NHC salts and complexes were tested and the IC50 values of these NHC salts and complexes were determined by a proliferation BrdU enzyme-linked immunosorbent assay (ELISA) against HeLa (Human cervix carcinoma), HT29 (human adenocarcinoma) and L929 (mouse fibroblast) cell lines. The IC50 values are in the range of 11 +/- 1-126 +/- 3 mu M show that all new Ag(I)-NHC complexes especially complex 2b demonstrated remarkable cytotoxic activity against HeLa, HT29 and L929 while 1a-e NHC salts are found to be inactive against HeLa, HT29 and L929. Also, the high IC50 value of complex 2b against L929 cells can be interpreted high selectivity against healthy cells. This complex has been highlighted as new types of metalodrug.
