Öğe Benzimidazol iskeletli, Pd-NHC içeren kataliz sisteminin Sonogashira tepkimelerindeki katalitik özelliklerinin ıncelenmesi(İnönü Üniversitesi, 2015) Yavuz, KemalFenilasetilen ve aril halojenürlerin bakır varlığında Pd komleksleri ile katalizlenen Sonogashira eşleşme tepkimesi organik sentezlerde önemli bir araçtır ve doğal bileşik sentezleri, biyolojik aktif moleküller ve mataryal bilimi gibi çeşitli alanlarda yaygın olarak kullanılmaktadır. Hammaddelerin maksimum kullanımı ve minimum atık madde oluşturulması kaygısı hem laboratuvarda hem de endüstride uygun katalizör ve çevre dostu sentez metodları için araştırmaları harekete geçirmiştir. Diğer taraftan kimyasal tepkimeleri gerçekleştirmek içinyeşil bir sentez metodu olarak bilinen mikrodalga ışıma kullanımı,son zamanlarda organik sentezlerede çok yönlü bir araç olmuştur. Hızlı ısıtma, tepkime süresini azaltma ve çoğu durumlarda ürün verimini artırma, mikrodalga ışınlamayı genel kullanılan bir araç yapmıştır. Bu bağlamda, Pd-NHC katalizörlerinin mikrodalga ile birlikte kullanımı, mikrodalga şartlarında sıcaklık meyilinin aşağı yönelmesi nedeniyle katalizör ömrünün uzayabilmesi, alışılmış ısıtma sistemlerine karşı önemli bir üstünlüğe sahip olabilecektir. Pd-NHC'lerin çeşitli C-C eşleşme tepkimeleri için etkin katalizör oldukları bilinmektedir ve bu tip katalizörler, Suzuki-Miyaura, Heck-Mizoroki ve Buchwald-Hartwig tepkimeleri ile karşılaştırıldığında Sonogashira tepkimesinde daha az sıklıkla kullanılmaktadır. Pd-NHCkatalizörlü Sonogashira tepkimelerinin veriminiartırmak amacıyla, bu tez kapsamında karben öncülü olarak yeni benzimidazol tuzları sentezlendi ve Sonogashira tepkimelerindeki katalitik özellikleri bakır nanopartikül varlığında mikrodalga ışınlama şartlarında araştırıldı. Benzimidazol tuzu, Pd(OAc)2, bakır nano partikül, baz ve çözgenden oluşan katalizör sisteminin katalitik etkinliği GC-MS tekniği kullanılarak seçilmiş tepkimelerde belirlendi.Katalizörsisteminin Sonogashira tepkimelerindeki optimum katalitik etkinliği mikrodalga ışınlama altında 10 dakikada elde edildi. Bu çalışmada sentezlenen tüm yeni benzimidazol bileşiklerinin yapısı 1H-NMR, {1H}-13C-NMR, FT-IRspektroskopik teknikleri ve element analizi ile aydınlatıldı. Katalizör olarak test edilen hem yeni (1-20) hemde bilinen (I-XX) bileşiklerin yapıları ve Sonogashira tepkimesi için optimum tepkime şartları aşağıda verilmiştir.Öğe Efficient and green catalytic system incorporating new benzimidazolium salts for the Sonogashira cross-coupling reaction(Wiley, 2018) Yavuz, Kemal; Kucukbay, HasanA number of novel benzimidazole salts were synthesized and their structures were determined using H-1 NMR, C-13 NMR and infrared spectroscopic techniques and elemental analysis. A catalyst system consisting of Pd(OAc)(2) and copper nanoparticles in the presence of Cs2CO3 and incorporating the novel benzimidazole salts in poly(ethylene glycol) solvent significantly improved the yields of Sonogashira reactions between aryl halides and phenylacetylene under microwave irradiation in 10min.Öğe Mikrodalga yardımıyla sübstitüye siklik diazo bileşiklerinin sentezi ve özelliklerinin incelenmesi(İnönü Üniversitesi, 2009) Yavuz, KemalOrganik sentezde mikrodalga yöntemi ; diğer geleneksel yöntemlerden hızlı ve temiz bir yöntem olduğu için organik kimya alanında büyük öneme sahiptir. Tezimizin temel amacı sübstitüye siklik diazo bileşiklerinin mikrodalga koşullarında sentezini gerçekleştirmek ve bu yöntemin üstünlüklerini ortaya koymaktır. Çalışmamız üç bölümden oluşmaktadır. Birinci bölümde başka araştırmacılar tarafından sübstitüye diazin bileşikleri üzerine yapılmış çalışmalar özetlenmiş ve organik kimyada mikrodalga sentez tekniğinin özellikleri açıklanmıştır. İkinci bölümde deneysel ayrıntılar verilmiştir. Üçüncü bölümde sonuç ve tartışmalar sunulmuştur. Deneysel çalışma iki kısımdan oluşmaktadır. Araştırmada önce 1-sübstitüye perimidin bileşikleri (1a-h) mikrodalga tekniği ile sentezlenip yapıları spektroskopik yöntemlerle aydınlatılmıştır. İkinci kısımda ise 1-sübstitüye perimidin türevleri (1) kullanılarak 1,3-disübstitüye perimidin tuzları (2a-f) hazırlanmıştır. Sentezlenen 1 bileşikleri alkil halojenürlerle etkileştirilerek mikrodalga koşullarında 2 tipi tuzlar elde edildi. 2 tipi bileşiklerin spektroskopik yöntemlerle yapıları aydınlatıldı. Deneysel çalışmalarımızı diazin bileşiklerinin önemli bir elemanı olan perimidinin mikrodalga ışınlama altında sentezi ve 1,3-disübstitüye bileşiklerinin sentezi ve yapılarının aydınlatılması oluşturmuştur.Öğe Synthesis and evaluation of anticancer properties of novel benzimidazole ligand and their cobalt(II) and zinc(II) complexes against cancer cell lines A-2780 and DU-145(Elsevier Science Sa, 2019) Yilmaz, Ulku; Tekin, Suat; Bugday, Nesrin; Yavuz, Kemal; Kucukbay, Hasan; Sandal, SuleymanEighteen new cobalt(II) or zinc(II) complexes of benzimidazole bearing 1-benzyl and 2-phenyl moieties were synthesized from the reaction of appropriate benzimidazole ligands and CoCl2 or ZnCl2. Their structural characterizations were done by IR, NMR (H-1, C-13) and UV-VIS spectrometers. Cytotoxic activities of eighteen new complexes and three benzimidazole ligands were determined using A-2780 (human ovarian) and DU-145 (human prostate) cell lines. Antitumor properties of all compounds were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Cell viability assay for the tested benzimidazole derivatives was performed and the LogIC(50) values of the compounds were calculated after a 24-hour treatment. All tested benzimidazole derivatives showed higher or comparable antitumor activity against A-2780 cell lines compared to the standard drug docetaxel with a LogIC(50) value of -0.81 mu M (p < 0.05). Eight of the examined compounds (1, 3, 5, 6, 7, 9, 10 and 13) showed high cytotoxic activity against A-2780 compared to the standard drug docetaxel. While the LogIC(50) of the docetaxel was -0.81 mu M for A-2780 cells at 24 h, the IC50 values of compounds 1, 3, 5, 6, 7, 9, 10 and 13 were - 0.97, -1.30, - 0.22, 0.13, - 0.16, - 0.73 and - 0.53 mu M, respectively. Three of the compounds 1, 18 and V showed high cytotoxic activity against DU-145 compared to docetaxel (p < 0.05). A mixture of the benzimidazole salts (1-5), Pd(OAc)(2), and K2CO3 in DMF-H2O catalyzed, in high yield, the Suzuki-Miyaura and the Heck-Mizoroki cross-coupling reactions assisted by microwave irradiation in 5 min. The novel benzimidazole salts (1-5), Pd(OAC)(2), CS2CO3, PEG, and Cu nanoparticles catalyzed, in high yield, the Sonogashira coupling reaction promoted by microwave irradiation in 10 min. The same benzimidazole salts (1-5), Pd(OAc)(2), CS2CO3, and TBAB catalyzed, in moderate or low yield, the Buchwald-Hartwig reaction assisted by microwave irradiation in 60 min. In this work, we present the synthesis of a series of 4-(methylthio)benzyl substituted benzimidazolium salts. All compounds were characterized using NMR (1H and 13C) and FTIR spectroscopic methods as well as an elemental analysis technique. The molecular and crystal structures of the compound 1a were determined by X-ray crystallography revealing that the compound crystallized in the trigonal space group R-3. Enzyme inhibition studies demonstrated that a new series of sulfurous compounds precursors were highly potent inhibitors for xanthine oxidase (XO) and acetylcholinesterase (AChE) enzyme. The IC50 values were found in the range of 0.548 +/- 0.033 to 0.725 +/- 0.043 mu M for XO promising strategy for the treatment from gout disease, while IC50 values were found in the range 0.813 +/- 0.076 to 1.149 +/- 0.072 mu M toward AChE as the key enzyme promising strategy for the treatment of neurological disorders such as Alzheimer's disease (AD). Furthermore, pharmacodynamics studies prove the binding interaction patterns, structural orientations and drug potential of sulfide derivatives in the binding sites of xanthine oxidase (XO) and acetylcholinesterase (AChE) enzymes. Potential inhibitors (compounds 1d-f) were compared with standard compounds allopurinol (for XO) and donepezil (for AChE). Compared to the positive compound of target XO, the 4-vinylbenzyl group of potential compound 1f and the 4-methylbenzyl group of compound le more effectively formed electrostatic and hydrophobic interactions with the target's interaction site. While donepezil as standard compound interacts only at the peripheral anionic site of AChE, the related compounds interact with both regions (PAS and CAS sites) of the same target. These compounds were placed at the active sites of the respective targets by molecular docking method using AutoDock software. Binding energy, binding modes and interaction types were used to evaluate the series of 4-(methylthio)benzyl substituted benzimidazolium salts's ability to bind to each target. Binding energy lower than zero remarks spontaneous binding, and equal and/or lower than -5 kcal/mol remarks good binding. Besides XO, the related compounds show higher activity against the AChE enzyme. They can also be analyzed as strong candidate compounds in biological studies of related enzymes.
