Öğe Analysis of E6/E7 mRNA gene expression by means of NucliSENS EasyQ (NASBA) technique in patients suffering from cervical dysplasia with HPV positive(7847050 Canada Inc, 2017) Ciplak, B.; Yilmaz, E.; Otlu, B.; Coskun, E. Materials and Methods: This study included 77 patients diagnosed with cervical dysplasia. The patients were grouped based on cervico-vaginal smear anomalies. Digene HC2 DNA Collection Device transport medium was used for taking and keeping cervical samples, QIA symphony SP device, and QIAsymphony DSP AXpH DNA kit were used for HPV DNA extraction from cervical samples kept in transport medium, HPV Q24 complete kit, RotorGene and PyroMark Q24 for detection of HPV-DNA and determination of HPV types and NucliSENS Easy Q Genetic Analyzer technique for determination of E6/E7 gene expression in HPV positive samples. SPSS (Statistical Package for Social Sciences) Version 22.0 was employed in statistical evaluation of the research date. Results: The most common HPV was considered as type 16 in this study. When pathology specimens of patients in whom dysplasia was detected in their cervical biopsies were examined in terms of HPV E6/E7 mRNA expression, a statistically significant difference was found between the normal and pathology groups (p < 0.05). Conclusion: Today, the presence of HPV infection in etiology of cervical smear pathologies and dysplasia is undisputedly accepted. The main determinant factor of HPV virulence is the frequency of E6 and E7 gene expressions existing in the DNA structure and responsible for virulence. NucliSENS_Ea_syQ (NASBA) is a technique employed to analyze mRNA gene expression in an accurate, reliable, and rapid way. M.; Kahraman, A.; Yilmaz, E.; Akatl, A.; Kural, H.OBJECTIVE: Detection of endometrial cancer (EC) is crucial in pre-operative EC management. The study aimed at determining the most reliable sequence in magnetic resonance imaging (MRI) evaluation at the EC. Different sequences were compared using histopathological results as the gold standard. PATIENTS AND METHODS: In this retrospective study, 62 women diagnosed with EC were examined using 3 Tesla MR and post-hysterectomy histopathological examination. The MRI protocol included diffusion- weighted imaging (DWI), T2 weighted images, Dynamic Contrast T1 weighted images, and ZOOMit DWI. Apparent diffusion coefficient (ADC) maps were also obtained. ADC maps were calculated for both DWI and ZOOMit DWI. All serial images were analyzed by endometrial distance to account for myometrial invasion and tumor size. All sequences and histopathological results were compared using the paired sample t-test. RESULTS: In all 32 women, post-hysterectomy histopathological confirmation was obtained. Mean myometrial invasion values were not significantly different after comparing DWI, ZOOM it DWI, and Dynamic contrast enhancement (DCE)-T1WI (p=0.054, p=0.039). The first strong correlation was found between DWI and ZOOMit DWI (0.943). The second was between ZOOMit DWI and DCE-T1 WI (0.949). Comparing each set of sequences, no statistically significant differences in tumor size ( p > 0.05) were found. Also, no statistically significant differences between images and histopathological size were found. The ADC values of ZOOMit DWI indicated that this method detected significant differences between EC histological subtypes. CONCLUSIONS: Each sequence is essential and valuable, whereas priorities may vary depending on the desired information. The most valuable sequences for the myometrial invasion were ZOOMit and conventional DWI. I.; Sahin, N.; Ciplak, B.; Ekici, K.Objective: Evaluation of mean platelet volume (MPV), neutrophil to lymphocyte ratio (NLR), and platelet count in the detection of malignant and benign ovarian tumors. Material and Methods: Sixty-six patients admitted to the present clinic over three years were included in the study. Patients diagnosed with ovarian cancer were grouped into the areas of epithelial and granulosa cell tumor diagnosis. The values were compared to patients with benign cystic structure. Patients' preoperative hematologic parameters and their values four weeks postoperatively were analyzed. Statistical analyses were performed with SPSS software. Results: MPV, NLR, and platelet count were observed at a higher rate as statistically significant in patients diagnosed with malignant ovarian cancer, compared to those with benign adnexal mass. I.; Gul, M.; Sahin, N.; Tuncay, G.; Simsek, Y.Objective: Examination of the role of nuclear factor-kappa beta (NF-kappa B) expression in the etiopathogenesis of endometrial cancer, by means of the immunohistochemical method. Materials and Methods: Patients who applied to participate in the study at the clinic were grouped into three categories: those diagnosed with benign endometrial pathology, those with endometrial hyperplasia, and those with endometrial cancer. NF-kappa B analysis was conducted in the endometrial tissues of the patients' paraffin blocks by means of the immunohistochemical method. For objective assessment purposes, the H score of each patient was calculated. SP SS 15.0 program was employed for statistical analysis. Results: The average H score of the first group, comprising benign endometrial pathologies, was 102.4 +/- 85.9, that of the hyperplasia group was 143.6 +/- 122.4, and that of the cancer group was 276.8 +/- 61.8. The average values of groups 1 and 2 were similar (p = 0.349); however, the third group's average H score was significantly higher (p < 0.001). Conclusion: NF-kappa B, which is a critical mediator in the inflammation process, might be related to the development of premalign and malign endometrial changes.Öğe Resveratrol successfully treats experimental endometriosis through modulation of oxidative stress and lipid peroxidation(Wolters Kluwer Medknow Publications, 2014) Yavuz, Simsek; Aydin, Nasuhi E.; Celik, O.; Yilmaz, E.; Ozerol, E.; Tanbek, K.Background and Aims: The purpose of this study was to investigate the potential therapeutic efficiency of resveratrol in the treatment of experimental endometriosis in rats. Settings and Design: Experimental study was carried out in a University hospital. Materials and Methods: Endometriosis was surgically induced in 24 female rats. Four weeks after this procedure, the viability and dimensions of the endometriosis foci were recorded. Rats were then randomly divided into three groups: (1) Control group (n = 8); (2) low dose (10 mg/kg) resveratrol group (n = 8); (3) high dose (100 mg/kg) resveratrol group (n = 8). At the end of the 7-day treatment, blood samples were taken and laparotomy was performed. The endometrial implants were processed for biochemical, histological and immunohistochemical studies. Statistical Analysis Used: The Kruskal-Wallis H test and one-way ANOVA test were used. Results: Resveratrol-treated rats showed significantly reduced endometriotic implant volumes (P = 0.004). After treatment, a significant and dose-dependent increase in activities of superoxide dismutase and glutathione peroxidase in serum and tissue of the rats in Group 2 and Group 3 was detected. Similarly, serum and tissue malonyl dialdehyde levels and tissue catalase levels were significantly higher in Group 3 than that of control animals. Histological scores and proliferating cell nuclear antigen expression levels were also significantly reduced in Group 2 and Group 3 than that of control group. Conclusion: In a rat endometriosis model, resveratrol showed potential ameliorative effects on endometriotic implants probably due to its potent antioxidative properties.Öğe Role of endometrial concentrations of heavy metals (cadmium, lead, mercury and arsenic) in the aetiology of unexplained infertility(Elsevier, 2014) Tanrikut, E.; Karaer, A.; Celik, O.; Celik, E.; Otlu, B.; Yilmaz, E.; Ozgul, O.Objective: To determine the role of endometrial concentrations of heavy metals (cadmium, lead, mercury and arsenic) in the aetiology of unexplained infertility. Study design: Thirty-three women with unexplained infertility and 32 fertile women were recruited. Endometrial biopsies were collected during the putative window of implantation (cycle days 20-24). The concentrations of cadmium, lead, mercury and arsenic were measured in endometrial biopsy specimens using atomic absorption spectrometry. Results: Cadmium was detected in 91% (30/33) of women with unexplained infertility, compared with 34% (11/32) of fertile women. The median endometrial cadmium concentration was 19.58 (interquartile range 1.46-30.23) mu g/l in women with unexplained infertility, compared with 0.00 (interquartile range 0.00-0.40) mu g/l in fertile women. Lead was detected in 15% (5/33) of women with unexplained infertility and 3% (1/32) of fertile women. Mercury and arsenic were not detected in any endometrial samples from either group. Conclusion: A significant difference in endometrial cadmium concentration was found between women with unexplained infertility and fertile women. This suggests that cadmium may be a contributing factor in the aetiology of unexplained infertility. (C) 2014 Elsevier Ireland Ltd. MATERIALS AND METHODS: Thirty-five patients with abnormal cervical cytology (study group) and 10 women with normal cytology (control group) were included in the study. The polymorphisms of XRCC1 Arg194Trp, XRCC1 Arg399Gln and XPD Lys751Gln genes were investigated from the blood samples. RESULTS: There was no statistically significant difference in allele frequencies of XPD gene among the groups (p = 0.097), while XRCC1R399Q gene polymorphism was strikingly more frequent in the study group than that of control cases (p = 0.029). The prevalence of XRCC1R194W gene polymorphism on the other hand, was similar between the groups (p = 0.579). CONCLUSIONS: Patients with abnormal and normal cervical cytology have similar XPD gene polymorphism. However, the frequency of gene polymorphism in XRCC1 Arg 399 Gln codon was significantly higher in abnormal cervical cytology group.
