Özet
An amperometric enzyme electrode for glucose determination was prepared by anodic polymerization of pyrrole and concomitant incorporation of glucose oxidase on Pt substrates in A KCl solution at a potential of 0.8 V vs. Ag/AgCl. The amperometric response to glucose was measured at a potential of 0.7 V in order to oxidize the hydrogen peroxide generated. Effects of various parameters (e.g., film thickness, concentrations of monomer, supporting electrolyte, and enzyme) on the electrode preparation and the effects of buffer concentration, temperature, and pH, on the chronoamperometric response of the prepared enzyme electrode were systematically investigated and the optimum values for each parameter were determined. If was found that a biosensor, with a response time of less than 5 s, can be easily prepared in ca. 5 min. Activation energy of the enzymatic reaction was calculated as 13 kJ/mol and the linearity of the enzyme electrode response ranged from 2.0 to 15 mM of glucose. The specificity of the enzyme electrode was tested for ascorbic, succinic and maleic acids, and sucrose, and no discernible signal was detected above the background current. Moreover, the response of the electrode was found to be stable for about one month.
