Öğe Assessment of BCL-2 expression in actinic keratosis, keratoacanthoma and seborrheic keratosis cases(2019) Onder, Sevda; Erdem, Havva; Turhan Haktanir, Nurten; Balta, Hilal; Akcay Celik, MuruvvetAim: One of the main functions of apoptosis is the elimination of cells with damaged DNA, including premalignant cells in skin. Apoptosis is regulated by the balance between antiapoptotic and proapoptotic proteins. The bcl–2 protein has been shown antiapoptotic effect. It protects cell against apoptosis induced by different death-inducing signals. Material and Methods: In this study the authors have analyzed imunohistochemically the expression of bcl-2 protein in the histopathological variants of the precancerous lesion [actinic keratosis (AK) (28 cases) and keratoacanthomas (KA) (12 cases) and seborrheic keratosis (SK) (22 cases) in skin. Results: In cases of AK, bcl-2 expression was confined to basal cell layer, as well as in two cases of thorough epidermis. SK expression of bcl–2 protein was in areas of basaloid proliferation.KA expression of bcl-2 protein was not dome-shaped keratin-filled crater areas but bcl-2 expression was confined of non crater areas. Conclusion: In conclusion, bcl-2 expression supports the observation that tumor cells are derived from basal keratinocytes in AK and SK and not support the observation that tumor cells are derived from basal keratinocytes in KA.Öğe Claudin-5 (tight junction) expression level changes in achilles tendon healing(2019) Cirakli, Alper; Cirakoglu, Derya; Erdem, Havva; Uzun, Erdal; Cankaya, SonerAim: This study aimed to reveal the relationship between changes in Claudin-5 expression and the duration of healing in Achilles tendon injury. Material and Methods: 18 Achilles tendons of Wistar-Albino rats were used in the study. Rats were divided into 3 groups as 6 rats in each group, group 1; sham group, group 2; tendon repair group (sacrificed after 3 weeks), group 3; tendon repair group (sacrificed after 6 weeks). Immunohistochemically, the tendons were stained with Claudin-5 and the degree of staining with light microscope was rated between 0 and 3. The obtained scores were compared with Kruskal Wallis test and Posthoc analysis. Results: The scores were 0.5 ± 1 (0-1) in group 1.1 ± 1 (1-2) in group 2 and 1.5 ± 1 (1-2) in group 3. A statistically significant difference was found between the groups (p = 0.026). In the posthoc analyzes, there was a significant difference between group 1 and 3, but there was no significant difference between groups 1 and 2 and between groups 2 and 3. Conclusion: The expression of claudins is regulated by many factors, including hormones, various cytokines, and epithelialmesenchymal transition-related transcription factors. In this study, the increase in the expression of Claudin-5 was noticed in proportion to the progress of primary wound healing. This relationship may be a part of the repair mechanism. The role of claudin levels in intercellular passage is crucial for function as it is important for cell signaling. Achilles tendon healing can be attributed to a laboratory parameter such as claudin. This can help to understand the recovery rate and can help early return to work or sport. We believe that as a laboratory parameter Claudin-5 may be useful in the evaluation of tendon healing.Öğe Investigation of survivin gene polymorphism in patients with gastric carcinoma(Dicle Medical Journal / Dicle Tip Dergisi, 2012) Yılmaz, Nesibe; Yaykaşlı, Kürşat Oğuz; Soğuktaş, Hatice; Oktay, Murat; Erdem, Havva; Kaya, Ertuğrul; Ekinci, Aysun; Kaya, SavaşAmaç: Dünya genelinde mide kanserinin insidansı düş- mesine rağmen hala önemli bir sağlık problemidir. Türkiye’de ise yılda 30.000 yeni mide kanseri vakasıyla 2. en sık görülen kanserdir. Mide kanseri genetik ve çevresel faktörlerin etkileşimiyle ortaya çıkan çok faktörlü karma- şık bir hastalıktır. Kanserli dokuda aşırı ifade edilen survivin, apoptozis inhibe edici proteinlerdendir. Bu çalışmada Survivin -31 G/C polimorfizmi ile mide kanseri arasındaki ilişki araştırıldı. Gereç ve yöntem: Çalışma Düzce Üniversitesi Araştırma ve Uygulama Hastanesi Patoloji Laboratuvarına gelen mide kanseri tanısı konmuş 46 hasta ve sağlıklı bireylerin oluşturduğu 42 kişilik kontrol grubu ile gerçekleştirildi. Bu bireylerin genotipi Düzce Üniversitesi, Tıp Fakültesi, Tıbbi Genetik Anabilim Dalı Laboratuvarlarında PCR-RFLP yöntemiyle tayin edildi. Bulgular: Hasta grubunda, GG genotipi 16 (% 34,8), GC genotipi 21 (% 45,7) ve CC genotipi ise 9 (% 19,6) olguda saptandı. Kontrol grubunda ise, genotip dağılımı sırasıyla 13 (% 31), 26 (% 61,9) ve 3 (% 7,1) bulundu. Hasta ve kontrol grubu karşılaştırıldığında istatistiksel olarak anlamlı bir fark saptanamadı. Fakat CC genotipine sahip bireylerin mide kanserine yakalanma riskinin GG (OR=1,52) daha fazla risk oluşturduğu bulundu. Sonuç: Bu çalışma bildiğimiz kadarıyla Türk toplumunda mide kanseri ile Survivin -31G/C polimorfizmini araştıran ilk çalışmadır. Elde ettiğimiz sonuçlar hasta ve kontrol gruplarımızın temsil ettiği toplum kesitinde mide kanseri ile Survivin -31 G/C polimorfizmi arasında anlamlı bir ilişki olmadığını göstermekle birlikte CC genotipinin mide kanserine yatkınlık oluşturduğu düşünülebilir.Öğe Meme kanserli hastalarda TIMP2 gen polimorfizmlerinin sıklığının araştırılması(EUROPEAN JOURNAL OF MEDICAL SCIENCE, 2012) Göksen Arslan, Ulviye; Özmerdivenli, Recep; Yaykaşlı, Kürşat Oğuz; Oktay, Murat; Erdem, Havva; Soğuktaş, Hatice; Yılmaz, Nesibe; Yaykaşlı, EmineThe breast cancers were placed in first rank among other cancer types in women, and second rank after lung cancer in terms of death in the World. It is estimated that this cancer has 20/100.000 frequency in east region and 40- 50/100.000 frequency west region according to present data in Turkey. Matrix Metalloproteinase (MMP), proteolytic enzymes involve in the breakdown of proteins located in the cell membrane and intercellular matrix such as collagen, elastin, proteoglycans, and gelatin. The tissue inhibitors of metalloproteinases (TIMPs) are proteins which are specific and natural inhibitor for MMPs. Its primary tasks regulate the activation of MMPs by suppress. The destruction of the balance between MMPs and TIMPs, may lead the emergence of pathological processes. In this study, the effect of TIMP2 (-418) G/C andTIMP2 (303) G/A polymorphisms in breast cancer patients in Turkish population were analyzed by Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP).The statistically significant difference was found between the control group and patients with breast cancer. However it is found that TIMP2 (303) GA genotype increases the risk of developing breast cancer at 1.486fold (p=0519), TIMP2 (-418) CC genotype increases the risk of developing breast cancer at 3.719-fold (p=0.519). In our knowledge, this study is the first to evaluate the relationship between breast cancer and TIMP2 gene polymorphisms in Turkish population. Our results show that there is no any association between breast cancer and TIMP2 gene polymorphisms in the community which is represented by our study and control groups. It was concluded from results that TIMP2 (303) GA genotype ile TIMP2 (-418) CC genotype may create the susceptibility to breast cancer.
