Öğe ASSESSMENT OF PHYTOTOXIC AND GENOTOXIC EFFECTS OF ANATASE TiO2 NANOPARTICLES ON MAIZE CULTIVAR BY USING RAPD ANALYSIS(Parlar Scientific Publications (P S P), 2018) Mutlu, Fatma; Yurekli, Fusun; Mutlu, Birol; Emre, Fatma Bilge; Okusluk, Funda; Ozgul, OnurIn this study, early phytotoxic and genotoxic effects of chemically synthesized nano-TiO2 (anatase) were investigated in seedlings of maize (Zea mays cv. Hido). Control (Hoagland) and NaCl (300 mM) groups were administered with nano-TiO2 (0.1%, 0.2% and 0.3%). Seed germination and seedling growth parameters of maize cultivar were markedly inhibited by salt stress. Nano-TiO2, without depending on concentration, was found to be ineffective on germination percentage in all administrations. On the contrary, nano-TiO2 caused significant increases in root-stem length and fresh-dry weights especially in NaCl+nano-TiO2 administrations. In salt administered samples, 0.3% nano-TiO2 increased root length 1.4 fold, stem length 4.8 fold, and fresh weight 1.2 fold. Genotoxic properties of nano-TiO2 in seedlings of maize were evaluated by using randomly amplified polymorphic DNA (RAPD-PCR). In RAPD analysis, 20 RAPD primers were found to produce unique polymorphic band profiles at different concentrations of nano-TiO2 maize seedlings. Genomic template stability (GTS), a qualitative measurement of changes in RAPD patterns of genomic DNA, decreased depending on increasing nano-TiO2, NaCl, and NaCl+nano-TiO2 concentrations. Therefore, it could be concluded that nano-TiO2 of appropriate dose can be used to ameliorate negative effects of salt stress by increasing growth potential of maize.Öğe Blood Cadmium Concentrations in Women with Ectopic Pregnancy(Humana Press Inc, 2018) Karaer, Abdullah; Tuncay, Gorkem; Tanrikut, Emrullah; Ozgul, OnurThe purpose of this study is to investigate the relationship between the blood level of cadmium and the occurrence of ectopic pregnancy. Forty-one (41) case patients with ectopic pregnancy and 41 uncomplicated intrauterine pregnant patients as controls were recruited. The concentrations of cadmium (Cd) were measured from blood samples using atomic absorption spectrometry. The cases and controls were similar in age, body mass index, and smoking habits. The median blood level of Cd was 0.32 mu g/l (interquartile range [IQR] 0.00-0.71) in the women with ectopic pregnancies and 0.34 mu g/l (IQR 0.09-0.59) in the controls. There was no significant association between blood cadmium levels and ectopic pregnancy.Öğe The effect of seminal plasma cadmium and lead levels on semen parameters in male subjects of infertile couples: a prospective cohort study(Taylor & Francis Inc, 2021) Tuncay, Gorkem; Karaer, Abdullah; Tanrikut, Emrullah; Ozgul, OnurThe aim of this prospective study was to investigate whether there is a relationship between seminal plasma cadmium (Cd) and lead (Pb) levels and semen parameters in male partners of infertile couples. Two hundred fifty one (251) men recruited with no history of occupational exposure to toxic metals. After semen analysis, seminal Cd and Pb levels were measured using atomic absorption spectrometry. Cadmium levels in men with hypospermia was significantly higher than men with normal semen volume (p = .049). In contrast, there was no statistically significant difference in median seminal Pb levels between men with hypospermia and men with normal semen volume (p = .13). There was no statistically significant association between seminal plasma Cd and Pb levels sperm concentration, motility, morphology and total progressively motile sperm count. These findings suggest that environmental Cd exposure may contribute to low semen volume in male partners of infertile couples. IMPACT STATEMENT What is already known on this subject? Toxic metals may adversely affect both male and female reproductive system. What the results of this study add? Seminal plasma cadmium levels in men with hypospermia were statistically significantly higher than men with normal semen volume. What the implications are of these findings for clinical practice and/or further research? Patients should be informed about possible adverse effects of toxic metals.Öğe Titanium dioxide-multiwalled carbon nanotube/polyimide composite film modified electrodes for simultaneous voltammetric detection of ascorbic acid, uric acid and dopamine as biomarker molecules(Springer, 2022) Gungor, Oznur; Ozgul, Onur; Aksoy, Busra; Okusluk, Funda; Koytepe, SuleymanTitanium dioxide-multiwalled carbon nanotube/polyimide (TiO2-MWCNT/PI) composite film modified electrodes for simultaneous detection of dopamine (DA), uric acid (UA), and ascorbic acid (AA) in clinical or biomedical applications were prepared. Firstly, TiO2-MWCNT and polyimide structures were synthesized. Then, TiO2-MWCNT/PI composites containing different ratios of MWCNT were subsequently prepared and characterized with chemical, thermal and morphological analysis techniques. Synthesized TiO2-MWCNT/PI composites were coated on the platinum electrode surface. Prepared electrodes were used to sense DA, UA, and AA with voltammetric measurements. LOD and LOQ parameters of modified electrodes for DA were observed 8.5114 mu M and 25.5342 mu M, respectively. The R-2 value for the DA in the linear range from 175 to 325 mu M was obtained 0.9967 (in the existence of AA and UA). After five repeatability results, 96.3% AA, 98.2% DA, and 88.3% UA peak currents were maintained. The modified electrode exhibited good selectivity, acceptable durability, high repeatability, and sensitivity.
