The presence of cannabinoid type 2 receptor (CB2), a member of the cannabinoid system, was detected in different regions of the brain. CB2 receptor plays a role in neuroinflammatory and neurodegenerative processes. We aimed to determine the possible effect of CB2 agonist JWH-133 in Okadaic acid (OKA)-induced neurodegeneration model mimicking Alzheimer's Disease (AD) through tau pathology. Materials and methods: In this study, 40 Sprague Dawley male rats were divided into 4 groups (Control, Sham, OKA, OKA + JWH-133). Bilateral intracerebroventricular (icv) injection of 200 ng OKA was performed in the OKA group. In the OKA + JWH-133 group, injection of JWH-133 (0.2 mg/kg) was performed intraperitoneally for 13 days different from the group of OKA. Morris water maze test was used to evaluate the spatial memory. Levels of caspase-3, phosphorylated tau (ser396), amyloid beta (A beta), tumor necrosis factor-alpha (TNF-alpha) and interleukin-1 beta (IL-1 beta) levels in brain cortex; and the hippocampus regions were examined by immunohistochemical methods. Key findings: In the OKA group, caspase-3, phosphorylated tau (ser396), A beta, IL-1 beta levels were higher in the cortex and hippocampus than in the other groups. The implementation of the JWH-133 reversed the increments in these parameters, and also prevented spatial memory impairment.Öğe Central irisin administration suppresses thyroid hormone production but increases energyconsumption in rats(Elsevıer ıreland ltd, elsevıer house, brookvale plaza, east park shannon, co, clare, 00000, ıreland, 2018) Tekin, Suat; Erden, Yavuz; Ozyalin, Fatma; Cigremis, Yilmaz; Colak, Cemil; Tekedereli, Ibrahim; Sandal, SuleymanIrisin, which is secreted from the skeletal muscle in response to physical exercise and defined as a thermogenic peptide, may play an important role in energy metabolism. Thyroid hormones, which are one of the other influential factors on the metabolic status, increase heat production and are the main regulators of energy metabolism. This study was conducted to determine the possible effects of irisin administration on thyroid hormones. Forty adult male Wistar albino rats were used in the study. The rats were equally divided into 4 groups (n = 10). The brain infusion kit was implanted in the groups, and irisin (or solvent as control) was centrally administered to the rats via osmotic mini pumps for 7 days. During the experiment, food consumption, body weights, and body temperatures of the animals were recorded. Food intake was significantly increased in the groups treated with irisin (p < 0.05), but their body weights were not changed. Hypothalamic TRH gene expression, serum TSH, fT3, and fT4 levels were significantly lower in the groups treated with irisin as compared to the naive and control groups (p < 0.05). In addition, irisin increased UCP1 mRNA expression in white and brown adipose tissue and UCP3 mRNA expression in muscle tissue in rats and also raised their body temperature (p < 0.05). Consequently, although central irisin administration has inhibitory effects on the hypothalamic-pituitary thyroid axis, it seems to be an important agent in the regulation of food intake and energy metabolism.Öğe Central Salusin-? Twenty-one non-smoking infertile men <40 years of age with no systemic or genetic disease were randomly selected from the pool of infertile couples applied to our clinic. Sperm samples were collected and evaluated according to WHO guidelines. DNA damage of sperm cells was assessed using neutral comet assay. Fifty cells per slide and two slides per sample were scored to evaluate DNA damage. The cells were visually classified into four categories based on DNA migration such as undamaged (UD), little damage (LD), moderate damage (MD) and significant damage (SD). Total comet scores (TCS) were calculated as: 1 x UD + 2 x LD + 3 x MD + 4 x SD. There was a negative correlation between the percentage of slow-and in situ-motion sperm cells in spermiograms and TCS (p < 0.001 and p < 0.05, respectively). The relationship between the percentage of non-motile sperm cells and TCS was negative (p < 0.001). This study provides new evidence that DNA damage in spermatozoa and sperm motility parameters are negatively correlated. We suggest that evaluation of sperm DNA by the neutral comet assay may be valuable to use in fertility research.Öğe Cytotoxic effect of endemic Tchihatchewia isatidea Boiss. from Turkey(Brill, 2021) Kirbag, Sevda; Keser, Serhat; Tekin, Suat; Inci, Sule; Sandal, SuleymanTchihatchewia isatidea Boiss (Brassicaceae) is known among the public as Reddish Bride, Paint and Bridal Flower. Flower parts are used for painting and flowering branches for treatment of cough. In this study, anticancer properties of water, ethanol and methanol extracts of Tchihatch-ewia isatidea flower parts were investigated against human breast (MCF-7), human colon (HCT-116) and human prostate (LNCaP) cancer cell lines. Changes in the viability of cancer cells were de-tected by utilizing 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method. Studies have shown that there is a statistically significant decrease in the cell viability of human breast cancer (MCF-7), colon (HCT-116) and prostate (LNCaP) cancer cells when extracts of T. isati-dea water, ethanol and methanol are used.Öğe Cytotoxic effects of coumarin substituted benzimidazolium salts against human prostate and ovarian cancer cells(Indian Acad Sciences, 2019) Karatas, Mert Olgun; Tekin, Suat; Alici, Bulent; Sandal, SuleymanCoumarin and benzimidazole derivatives have individual biological activities including anticancer. In this study, we aimed to synthesize coumarin-benzimidazole hybrids in order to investigate their anticancer properties. For this purpose, six 6-substituted-4-chloromethylene coumarin derivatives were synthesized. Sixteen coumarin substituted benzimidazolium chlorides were synthesized by the reaction of 4-chloromethylene coumarin and N-benzylbenzimidazole derivatives. All of the synthesized compounds were characterized by 1H and 13C NMR, IR spectroscopic techniques and elemental analyses. Cytotoxicities of all compounds were tested by [ 3-(4,5-dimethylthiazole)-2-yl]-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay against human prostate (PC-3) and ovarian (A2780) cancer cells. All compounds performed significant cytotoxicities at 100 mu Magainst both cancer cell lines. Moreover, some compounds performed significant activities at 1 mu M against both cancer cell lines and the obtained results suggest that this type of compounds are promising candidates for the treatment of human prostate and ovarian cancers.Öğe Cytotoxic potential of silver, palladium, rhodium, ruthenium, and iridium complexes of a cycloheptyl-substituted lipophilic N-heterocyclic carbene ligand(Taylor & Francis Ltd, 2023) Karatas, Mert Olgun; Sekerci, Guldeniz; Tekin, Suat; Sandal, Suleyman; Kucukbay, HasanLipophilicity is a crucial parameter for cytotoxicity of metal complexes, in many cases associated with increased activity. In the present study, we synthesized a series of N-heterocyclic carbene (NHC) complexes of different metals with a lipophilic benzimidazolium chloride to investigate and compare their cytotoxicity. Rh- (4), Ru- (5), and Ir-NHC (6) complexes of a cycloheptyl-substituted benzimidazole-based NHC ligand (1) have been prepared by transmetalation reaction between Ag-NHC and the corresponding metal compound. The newly synthesized complexes have been characterized by NMR, IR, LC-MS spectroscopy, and elemental analyses. The anti-growth effects of the newly synthesized Rh-, Ru-, and Ir-NHC complexes and previously reported NHC precursor (1), Ag- (2), and Pd-NHC (3) complexes against human breast (MCF-7), colorectal (Caco-2), ovarian (A2780), and prostate (LNCaP) cancer cell lines have been investigated by MTT assay. The results revealed that the compounds provide stronger anti-growth effects against all cell lines compared to standard drug cisplatin. The synthesized compounds were characterized by elemental analysis, FT-IR and NMR. DFT was used to study the quantum chemical calculations of geometries and vibrational wave numbers of 3-hydroxynaphthyl and p-tolyl substituted 1,2,4-triazoles in the ground state. The scaled harmonic vibrational frequencies obtained from the DFT method were compared with those of the FT-IR spectra and found good agreement. The synthesized 1,2,4-triazole-naphthyl hybrids were screened for the anticancer activity against MCF-7 breast cancer lines. Among them compounds 3 and 7 showed broad spectrum anticancer activity with IC50 values 9.7 mu M and 7.10 mu M, respectively and their activity is comparable to that of the standard drugs.
