Fenetil izotiyosiyanatın ratlarda yüksek yağ diyeti ve streptozotosin ile indüklenen yağlı karaciğer hastalığına etkilerinin incelenmesi

Özet
Amaç: Bu çalışmada, yüksek yağ dieti ve streptozotosin ile oluşturulan deneysel diabetin indüksiyonu sonucunda ortaya çıkan yağlı karaciğer hastalığında, oksidatif stres ve karaciğer hasarına karşı fenetil izotiyosiyanat'ın, SIRT1-Nrf2 yolağı üzerinden etkilerinin araştırılması amaçlanmıştır. Materyal-Metot: 30 adet (150-250g, 6-8 haftalık, erkek) Wistar Albino sıçanlardan her grupta 10 sıçan olacak şekilde üç grup halinde gruplandırıldı: Kontrol, yüksek yağlı diyet + Streptozotosin (35mg / kg), yüksek yağlı diyet + Streptozotosin (35mg / kg) + Fenetil izotiyosiyanat (40 mg / kg). Serum ALT seviyeleri ve serum lipidleri (TG, TC, HDL ve LDL / VLDL), antioksidan enzim aktiviteleri (SOD, CAT, GPx), MDA, GST, SIRT1 ve NF-ƙβ seviyeleri, ayrıca Nrf2'nin DNA bağlanma seviyeleri belirlendi. Karaciğer dokularında histolojik incelemeler yapıldı. Bulgular: Kontrol grubuna göre YYD+STZ grubunda, serum HDL düzeylerinin, karaciğer dokusunda antioksidan enzim aktivitelerinin (SOD, CAT, GPx), GST, SIRT1 and NF-ƙB miktarının, aynı zamanda Nrf2'nin DNA'ya bağlanma aktivitesinin azaldığı, serum ALT, serum lipid (TG, TC, LDL/VLDL) ve MDA düzeylerinin ise arttığı gözlenmiştir (p<0.05). PEITC verilen gruptaki bu parametrelerde düzelmenin olduğu belirlenmiştir (p<0.05). Histolojik incelemeler biyokimyasal analizleri desteklemiştir (p<000.1). Sonuç: PEITC, SIRT1/Nrf2 yolağı üzerinden YYD ve STZ ile indüklenen NAFLD'yi iyileştirmiştir. PEITC'nin NAFLD'ye karşı gelecek vaat eden ve etkili bir aday ajan olabileceğini söyleyebiliriz. Aim: In this study, it was aimed to investigate the effects of phenethyl isothiocyanate on the SIRT1-Nrf2 pathway against oxidative stress and liver damage in fatty liver disease caused by the induction of experimental diabetes induced by high fat diet and streptozotocin. Materials and Methods: 30 Wistar Albino rats (150-250g, 6-8 weeks old, male) were divided into three groups (each of 10 rats): control, high fat diet+ Streptozotocin (35mg / kg), high fat diet+ Streptozotocin (35mg / kg) + Phenethyl isothiocyanate (40mg / kg). Levels of serum ALT, and serum lipids (TG, TC, HDL, and LDL / VLDL), activities of antioxidant enzymes (SOD, CAT, GPx), and levels of MDA, GST, SIRT1, and NF-ƙβ, also activity of DNA binding levels of Nrf2 were determined. Histological examinations were performed on the liver tissues. Results: Compared to the control group, in the rats of the HFD+STZ group, serum HDL level, hepatic antioxidant enzyme activities (SOD, CAT, GPx), levels of GST, SIRT1, and NF-ƙβ, as well as DNA binding activity of Nrf2 decreased, serum ALT, serum lipids (TG, TC, LDL / VLDL) and MDA levels were observed to increase (p <0.05). It was determined that there was an improvement in these parameters in the group of PEITC treatment (p <0.05). Histological examinations supported biochemical analysis (p <000.1). Conclusion: PEITC improved NAFLD induced by HFD and STZ through the SIRT1-Nrf2 pathway. We can say that PEITC could be a promising and effective candidate agent against NAFLD.
