Özet
Amaç: Bu çalışmanın amacı resveratrolün meme kanseri hücre hattında hepsidin/ferroportin aksına etkisini değerlendirmektir. Materyal ve Metot: Çalışmada resvertrolün MDA-MB-231 hücre hattındaki sitotoksik dozunun belirlenmesi için MTT testi yapıldı. Resveratrolün MDA-MB-231 hücre hattında Hepsidin/Ferroportin aksına etkisini değerlendirmek için bu proteinlerin miktarlarına ELİSA ve Western Blot analizi yöntemi ile bakıldı. ELİSA testinde proteinlerin seviyeleri bağımsız iki örnekleme t-testi ile değerlendirildi. Proteinlerin miktarlarını gösteren Western Blot analizi görüntüleri İmage J programı ile analiz edildi ve istatiksel değerlendirmeler GraphPad Prism programı ile yapıldı. Bulgular: Resveratrolün MDA-MB-231 agresif meme kanseri hücre hattında IC50 konsantrasyonu 146.5 ?M olarak hesaplandı. Hem ELİSA testi hem de Western Blot analizinde kontrol grubuna kıyasla resveratrol uygulanan grupta hepsidin proteinin miktarı azalırken, ferroportin proteininin miktarı artmış olarak bulundu. Sonuç: Bu çalışmanın sonucunda, resveratrolün meme kanseri tıbbi tedavisine destek olacak bir tedavi olma ihtimali olduğu, fakat kesin önerilerde bulunmak için daha ileri in vitro, in vivo ve klinik çalışmalara ihtiyaç duyulduğu sonucuna varıldı.
Aim: The aim of this study was to evaluate the effect of resveratrol on the hepsidin/ferroportin axis in a breast cancer cell line. Material and Methods: In this study, MTT assay was performed to determine the cytotoxic dose of resveratrol in MDA-MB-231 cell line. To evaluate the effect of resveratrol on the Hepsidin/Ferroportin axis in the MDA-MB-231 cell line, the levels of these proteins were examined by ELISA and Western Blot analysis. The levels of proteins in ELISA assay were evaluated by independent two-tailed t-test. Western Blot analysis images showing the levels of proteins were analyzed with Image J program and statistical evaluations were performed with GraphPad Prism program. Results: The IC50 concentration of resveratrol in the MDA-MB-231 aggressive breast cancer cell line was calculated as 146.5 ?M. In both ELISA test and Western Blot analysis, the amount of hepcidin protein was found to be decreased and the amount of ferroportin protein was found to be increased in the resveratrol-administered group compared to the control group. Conclusion: As a result of this study, it was concluded that resveratrol has the potential to be a supportive therapy for breast cancer medical treatment, but further in vitro, in vivo and clinical studies are needed to make definitive recommendations.
