Özet
Amaç: Bu çalışmanın amacı, Malatya İnönü Üniversitesi Turgut Özal Tıp Merkezi Doku Tipleme Laboratuvarında 2020 yılında doku tiplemesi çalışılan hasta ve donörlerin Yeni Nesil Sekanslama (Next Generation Sequencing-NGS) yöntemi ile HLA allel ve haplotip frekanslarının belirlenmesidir. Materyal ve Metod: Malatya İnönü Üniversitesi Turgut Özal Tıp Merkezi Doku Tipleme Laboratuvarında 2020 yılında doku tiplemesi çalışılan 1081 kişiden oluşan hasta ve donörlerin Yeni Nesil Sekanslama (Next Generation Sequencing-NGS) yöntemi ile HLA allel ve haplotip frekanslarının belirlenmesi amacıyla çalışma gurubuna dahil edilmiştir. Yeni Nesil Sekanslama (Next Generation Sequencing) yöntemi ile bireylerin Human Leucocyte Antigeni allel ve haplotip frekansları belirlenmiştir. Bulgular: Çalışmada HLA*A, HLA*B, HLA*C', HLA*DP, HLA*DQ, HLA*DRB1'de en sık görülen alleller tespit edilerek HLA*A, HLA*B, HLA*C, HLA*DP, HLA*DQB1 ve HLA*DRB1 genindeki haplotip frekansları incelenmiştir. Sonuç: Çalışma sonucunda HLA*A'da en sık görülen üç allel 2402, 0201 ve 1101, HLA*B'da en sık görülen üç allel 5101, 3501 ve 3503, HLA*C'da en sık görülen üç allel 0401, 0701 ve 1203, HLA*DP'de en sık görülen üç allel 0401, 0201 ve 0402, HLA*DQ'da en sık görülen üç allel 0301, 0501 ve 0201, HLA*DRB1'de en sık görülen üç allel 1104, 0301 ve 1101 olarak tespit edilmiştir. Ayrıca HLA*A genindeki haplotip frekansı 0,403, HLA*B genindeki haplotip frekansı 0,333, HLA*C genindeki haplotip frekansı 0,409, HLA*DP genindeki haplotip frekansı 0,705, HLA*DQB1 genindeki haplotip frekansı 0,469, HLA*DRB1 genindeki haplotip frekansı 0,331 bulunmuştur.
