Abstract
Öz: Bu yazıda birçok hastanedeki hastane infeksiyonlarından sıklıkla izole edilen Acinetobacter baumannii, Escherichia coli ve Klebsiella türlerinin alttiplendirmesinde kullanılmak üzere optimize edilmiş, hızlı ve ortak bir “pulsed-field” jel eletroforez (PFGE) protokolü sunulmuştur. Optimize edilen yöntem basit, tekrarlanabilir ve farklı bakteriler için kullanıma uygun bulunmuştur. Ayrıca süreyi kısaltması, kullanılan çözelti ve enzimlerin düşük hacimleriyle çalışılabilmesi nedeniyle, ekonomik olarak değerlendirilmiştir. Çalışılan bakterilere bağlı salgınları değerlendirme ve hastane infeksiyonlarının yaygınlık derecesi hakkında yararlı bilgiler sunma potansiyeli vardır. Optimize edilen bu protokol, farklı merkezlere ait PFGE sonuçlarının birbirleriyle karşılaştırmasına olanak sağlayabilir. Başlık (İngilizce): A Rapid pulsed-field gel electrophoresis (PFGE) protocol developed for subtyping acinetobacter baumannii, escherichia coli, and klebsiella spp. Öz (İngilizce): This paper provided information about a rapid pulsed-field gel electrophoresis (PFGE) protocol optimized for subtyping of Acinetobacter baumannii, Escherichia coli, and Klebsiealla spp. which are commonly isolated from nosocomial infections in many hospitals. The procedure was simple, reproducible and versatile. Furhermore it is likely cost-effective because of reducing time, reagent volumes and enzymes. It has potential to identify outbreaks and follow spreading degree of nosocomial infections caused by the tested bacterial strains. This optimized protocol can allow PFGE fingerprinting profiles of the isolates from different settings to be compared.
