Öğe Acute physiological changes in elite free-style wrestlers during a one-day tournament(Edizioni Minerva Medica, 2016) Kafkas, Muhammed E.; Taskiran, Celal; Kafkas, Armagan Sahin; Ozen, Gokmen; Taskapan, Cagatay; Ozyalin, Fatma; Skarpanska-Stejnborn, AnnaBACKGROUND: The purpose of this study was to examine tree radical production, muscle damage and inflammation responses of well-trained wrestlers to a simulated one-day tournament of free-style wrestling. METHODS: Twelve elite competitive wrestlers with mean age (+/- SD) of 24.09 +/- 6.20 years, body mass 74.09 +/- 11.50 kg, and body height 174.90 +/- 8.8 cm and who had competed for national teams completed five matches according to the official Olympic wrestling tournament regulations. Blood sampling was collected before and after fifth match. Baseline blood testing was measured at 16:00 a.m. and then matches started at 12:00. Each match was implemented within one hour. Also, the resting time was 45 minutes following each match. The measurements were analyzed by Wilcoxon Signed Ranks Test, which is used to test for significant differences between pre- and post-test. RESULTS: The post-match lactate dehydrogenase creatine kinase (CK), and interleukin (IL)-6 levels were significantly increased compared with the baseline status. However. baseline malondialdehyde levels were not found significantly different compared with post-match. CONCLUSIONS: The current study ensured that one-day Free-Style wrestling tournament brings about significantly increasing on CK, LDH of muscle damage markers. Also, inflammatory status showed a progressive worsening during the course of one-day tournament. The study showed enhanced muscle damage markers and inflammatory status after one-day Free-Style wrestling tournament. Larazotide acetate (LA) is a molecule that acts as a tight junction (TJ) regulator by blocking zonulin (Zo) receptors in the intestine. Aims In our study, we aimed to investigate the effects of LA on intestinal barrier dysfunction and bacterial translocation in the AP model in rats. Methods Thirty-two male Sprague-Dawley rats were divided into 4 groups; control, larazotide (LAR), AP, and AP + LAR. The AP model was created by administering 250 mg/100 g bm L-Arginine intraperitoneally 2 times with an hour interval. AP + LAR group received prophylactic 0.01 mg/mL LA orally for 7 days before the first dose of L-Arginine. For intestinal permeability analysis, fluorescein isothiocyanate-dextran (FITC-Dextran) was applied to rats by gavage. The positivity of any of the liver, small intestine mesentery, and spleen cultures were defined as bacterial translocation. Histopathologically damage and zonulin immunoreactivity in the intestine were investigated. Results Compared to the control group, the intestinal damage scores, anti-Zo-1 immunoreactivity H-Score, serum FITC-Dextran levels and bacterial translocation frequency (100% versus 0%) in the AP group were significantly higher (all p < 0.01). Intestinal damage scores, anti-Zo-1 immunoreactivity H-score, serum FITC-Dextran levels, and bacterial translocation frequency (50% versus 100%) were significantly lower in the AP + LAR group compared to the AP group (all p < 0.01). Conclusions Our findings show that LA reduces the increased intestinal permeability and intestinal damage by its effect on Zo in the AP model in rats, and decreases the frequency of bacterial translocation as a result of these positive effects.Öğe An antibody of TNF-alpha did not prevent thioacetamide-induced hepatotoxicity in rats(Sage Publications Ltd, 2011) Demirel, Ulvi; Harputluoglu, Murat M. We investigated the effects of infliximab (a TNF-alpha antibody) on liver damage in thioacetamide (TAA)-induced hepatotoxicity in rats. Group 1 (n = 8) was the control group. In group 2 (n = 8), the TAA group, the rats received 300 mg/kg intraperitoneal (ip) TAA daily for 2 days. In group 3 (n = 8), the TAA + Infliximab (INF) group, infliximab (5 mg/kg ip daily) was administered 48 hours before the first dose of TAA daily for 2 days and was maintained for 4 consecutive days. In group 4 (n = 8), the INF group, the rats received only ip infliximab (5 mg/kg) daily. Livers were excised for histopathological and biochemical tests (thiobarbituric-acid-reactive substances [TBARS], and myeloperoxidase [MPO]). Serum ammonia, aspartate transaminase (AST), alanine transaminase (ALT), TNF-alpha, liver TBARS and MPO levels, and liver necrosis and inflammation scores in the TAA group were significantly higher than in the control and INF groups (all p < 0.01). All parameters except AST were not significantly different between TAA and TAA + INF. Methods: This study included 40 obese (26 females and 14 males, mean age 51.9 years) and 40 normal weight subjects (23 females and 16 males, mean age 53.5 years) with normal coronary angiograms. All the study participants underwent RT3DE to assess LA volume and mechanical function. Plasma NT-proANP was determined by ELISA method. Results: There was no significant difference between groups in left ventricular (LV) diameters and ejection fraction, which reflect LV systolic function. However, transmitral deceleration time, isovolumetric relaxation time, and peak late diastolic tissue Doppler velocity values, which reflect LV diastolic function, were found to be significantly higher in obese subjects when compared with controls. LA maximum volume (LAVmax), LAVmax index (LAVI), LA minimal volume (LAVmin), before atrial contraction volume (LAVpreA), LA active emptying volume, LA total emptying volume, and LA active emptying fraction, which reflect LA reservoir and pump functions, were also higher in obese subjects when compared with controls. LA passive emptying fraction was significantly lower in obese subjects than in controls. NT-proANP levels were similar between groups. There were positive correlations between NT-proANP level and LAVI, LAVmax, LAVmin, LAVpreA, and LA total and active emptying volumes. Conclusions: Left atrial mechanical functions and volumes are impaired in obese subjects. It is reported that there is a strong relationship between the nitric oxide (NO) level and the clinical prognosis of the infection in infections of G. intestinalis. The aim of the study was to investigate serum NO levels in samples taken from patients with G. intestinalis detected in the microscopic stool examination and from the control group consisting of healthy people and the presence of any relationship with Giardiosis. The study consisted of 60 patients diagnosed with G. intestinalis in microscopic stool examination, [31 (51.7%) male, 29 (48.3%) female] and 60 healthy people [28 (46.7% male, 32 (53.5%) female] as the control group. All patients in the patient group with gastrointestinal symptoms had chronic diarrhea and cysts and trophozoites were observed in microscopic examination of the feces in all of them. Measurement of serum NO level was performed using Cortas method. When the data were evaluated by T-test in the Independent Groups, NO levels in the patient group was found low at a statistically significant level. In Giardiosis, the decline in the NO level suggested that the disease causes a more severe clinical presentation. In addition to this, we consider it necessary that extensive experimental or clinical studies be conducted on the subject.Öğe Association of folic acid receptor ? in maternal serum with neural tube defects(Informa Healthcare, 2014) Celik, Ebru; Karaer, Abdullah; Turkcuoglu, Ilgin; Turhan, Ugur; Gungoren, Arif; Taskapan, Cagatay; Ozyalin, FatmaObjective: To evaluate whether serum folic receptor alpha levels are changed in women whose previous pregnancies were complicated with neural tube defects (NTDs). Methods: This was a case-control study that included 41 women as the control group who had previously had at least one healthy pregnancy and 37 women as the study group who had a previous pregnancy complicated with NTDs. Blood samples were obtained from all of the participants six weeks after the termination of pregnancy or delivery of a baby. Serum folate receptor alpha concentrations were analyzed using a commercially available enzyme-linked immunosorbent assay (ELISA) kit. Results: The mean concentrations of serum folate receptor alpha were significantly lower in the NTD cases compared to those in the control group (p = 0.02). There was no significant difference in mean serum folate titers between the NTD cases and the control group (p = 0.07). Conclusion: Low serum folic acid receptor alpha levels in the current study did not appear to be a regulatory marker of maternal folate homeostasis per se but rather a factor that contributed to the development of NTDs.Öğe Central irisin administration suppresses thyroid hormone production but increases energy consumption in rats(Elsevier Ireland Ltd, 2018) Tekin, Suat; Erden, Yavuz; Ozyalin, Fatma; Onalan, Ebru Etem; Cigremis, Yilmaz; Colak, Cemil; Tekedereli, IbrahimIrisin, which is secreted from the skeletal muscle in response to physical exercise and defined as a thermogenic peptide, may play an important role in energy metabolism. Thyroid hormones, which are one of the other influential factors on the metabolic status, increase heat production and are the main regulators of energy metabolism. This study was conducted to determine the possible effects of irisin administration on thyroid hormones. Forty adult male Wistar albino rats were used in the study. The rats were equally divided into 4 groups (n = 10). The brain infusion kit was implanted in the groups, and irisin (or solvent as control) was centrally administered to the rats via osmotic mini pumps for 7 days. During the experiment, food consumption, body weights, and body temperatures of the animals were recorded. Food intake was significantly increased in the groups treated with irisin (p < 0.05), but their body weights were not changed. Hypothalamic TRH gene expression, serum TSH, fT3, and fT4 levels were significantly lower in the groups treated with irisin as compared to the naive and control groups (p < 0.05). In addition, irisin increased UCP1 mRNA expression in white and brown adipose tissue and UCP3 mRNA expression in muscle tissue in rats and also raised their body temperature (p < 0.05). In this study, 40 male Sprague-Dawley rats were used (four groups; n=10). Apelin-13 at three different dosages (1, 5 and 50g/kg) was given intraperitoneally while the control group received vehicle the same route for a period of 14 days. In results, apelin-13 caused significant decreases in serum testosterone, luteinizing hormone and follicle-stimulating hormone levels (p<0.05). Administration of apelin-13 significantly increased body weights, food intake, serum low-density lipoprotein and total cholesterol levels (p<0.05), but caused significant decreases in high-density lipoprotein levels (p<0.05). Serum glucose and triglyceride levels were not significantly altered by apelin-13 administration. Significant decreases in both uncoupling protein (UCP)-1 levels in the white and brown adipose tissues and UCP-3 levels in the biceps muscle (p<0.05) were noted. Control group (S) rats were subjected to sham experimental hemorrhagic shock after which they underwent a sham operation, while trauma group (T) rats were subjected to hemorrhagic shock and subsequent head trauma with no treatment. Among the rats subjected to hemorrhagic shock and subsequent head trauma, those given 3% NaCl after this were named as HS group; those given HyperHeas [7.2% NaCl / 6% poly (O-2-Hydroxyethyl) starch] were named as HyperHS group; those given 0.9% NaCl were determined as NS group and those given ringer lactate as RL group. 24-hours later, the fluids’ effects were evaluated. The brain fluid content and INR levels were significantly higher in all the experimental groups when set against those in the NS group (p ranged <0.01 to <0.001). aPTT was significantly longer in the T and HS groups than in that seen in the NS group (p<0.001 for each). Rats in the HS and RL groups showed significantly more bleeding than those in the NS group (p<0.05). Of the treatment groups, the HyperHS group had more brain edema when compared to the NS and RL groups (p<0.05). The proportion of red neuron and necrosis was partially decreased in the treatment groups, with no significant difference determined between the HS, HyperHS, NS and RL groups (p>0.05). This study was conducted to determine the possible effects of irisin on nutritional behaviour. In this study, 40 male Wistar Albino rats were separated into 4 groups (n =10 for each group). Osmotic mini-pumps were connected to metal cannulas implanted to lateral ventricle; and artificial cerebrospinal fluid (vehicle), and 10 and 100 nM of irisin was infused for 7 days. The daily food and water consumptions and body weights of rats were followed up. After the infusion, the animals were killed, and the hypothalamus and blood samples were collected. NPY, POMC, and UCP2 mRNA levels in the hypothalamus were examined by RT-PCR. In serum, leptin and ghrelin levels as well as the levels of metabolic parameters were measured by using ELISA. It was determined that irisin administration increased the daily food consumption (p < 0.05), without causing significant changes in water consumption and body weight. Irisin also caused increases in ghrelin level in circulation and NPY and UCP2 mRNA levels in the hypothalamus, whereas it decreased the leptin level in circulation and POMC mRNA levels in the hypothalamus (p < 0.05). Otherwise, irisin caused decrease in LDL, triglycerides and cholesterol levels, while increasing HDL and glucose levels (p < 0.05). Results indicates that long-term irisin treatment increases food intake without increasing body weight associated with increased ghrelin, NPY and UCP2 mRNAs, and decreased leptin and POMC mRNA in the hypothalamus. Forty male Wistar Albino rats were divided into 4 groups with 10 animals in each group: control, sham, 10 and 100 mu M MOTS-c. Hypothalamus, blood, muscle, adipose tissues samples were collected for thyrotropin-releasing hormone (TRH), UCP1 and UCP3 levels were determined by the RT-PCR and western blot analysis. Serum thyroid hormone levels were determined by the ELISA assays. MOTS-c infusion was found to increase food consumption but it did not cause any changes in the body weight. MOTS-c decreased serum TSH, T3, and T4 hormone levels. On the other hand, it was also found that MOTS-c administration increased UCP1 and UCP3 levels in peripheral tissues. The findings obtained in the study show that central MOTS-c infusion is a directly effective agent in energy metabolism.
